Some water bodies in the Sinos River Basin (SRB) have been suffering the effects of pollution by residential, industrial and agroindustrial wastewater. The presence of cytotoxic and genotoxic compounds could compromise the water quality and the balance of these ecosystems. In this context, the research aimed to evaluate the genotoxicity and cytotoxicity of the water at four sites along the SRB (in the cities of Santo Antônio da Patrulha, Parobé, Campo Bom and Esteio), using bioassays in fish and cell culture. Samples of surface water were collected and evaluated in vitro using the Astyanax jacuhiensis fish species (micronucleus test and comet assay) and the Vero lineage of cells (comet assay and cytotoxicity tests, neutral red -NR and tetrazolium MTT). The micronucleus test in fish showed no significant differences between the sampling sites, and neither did the comet assay and the MTT and NR tests in Vero cells. The comet assay showed an increase in genetic damage in the fish exposed to water samples collected in the middle and lower sections of the basin (Parobé, Campo Bom and Esteio) when compared to the upper section of the basin (Santo Antônio da Patrulha). The results indicate contamination by genotoxic substances starting in the middle section of the SRB.Keywords: water quality, micronucleus test, comet assay, fish bioassays, cell culture. Avaliação da genotoxicidade e da citotoxicidade de amostras de água da Bacia do Rio dos Sinos, sul do Brasil ResumoAlguns corpos d'água da Bacia Hidrográfica do Rio dos Sinos (BHRS) vêm sofrendo os efeitos da poluição por efluentes domésticos, industriais e agroindustriais. A presença de compostos citotóxicos e genotóxicos pode comprometer a qualidade da água e o equilíbrio desses ecossistemas. Neste contexto, o objetivo do trabalho foi avaliar a genotoxicidade e a citotoxicidade da água em quatro pontos ao longo da BHRS (Santo Antônio da Patrulha, Parobé, Campo Bom e Esteio), utilizando bioensaios em peixes e em cultura celular. As amostras de água de superfície foram coletadas e avaliadas in vitro utilizando a espécie de peixe Astyanax jacuhiensis (teste de micronúcleo e ensaio cometa) e a linhagem celular tipo Vero (ensaio cometa e os testes de citotoxicidade vermelho neutro -VN e tetrazólio MTT). O teste de micronúcleos em peixes não apresentou diferenças significativas entre os pontos de coleta, assim como o ensaio cometa e os testes VN e MTT nas células Vero. O ensaio cometa demonstrou aumento nos danos genéticos em peixes expostos às amostras de água coletadas nos trechos médio e inferior da bacia (Parobé, Campo Bom e Esteio) em relação ao trecho superior da bacia (Santo Antônio da Patrulha). Os resultados indicam contaminação por substâncias genotóxicas a partir do trecho médio da BHRS.Palavras-chave: qualidade da água, teste de micronúcleo, ensaio cometa, bioensaios com peixes, cultura celular.

This research aimed to evaluate the air quality, by determining the concentrations of PM 2.5-10 , PM 2.5 and the metallic elements Al, Ba, Cd, Cr, Cu, Fe, Mn, Ni, Pb, Zn and Hg in the leaf part of ryegrass (Lolium multiflorum) in an area close to Schmidt Stream, at the lower section of Sinos River Basin (SRB), in a research campaign of six months, from October 2013 to March 2014. The particles collected in the PM sampling were analyzed by Scanning Electron Microscopy (SEM) combined with Energy Dispersive X-ray Spectrometry (EDS), in order to study their morphology and chemical composition. The mean concentration of PM 2.5-10 was 9.1 µg m . Concentrations of metallic elements, especially Pb, Cr and Zn, were classified as Class 4 (very high pollution levels), according to the classification proposed by Klumpp et al. (2004). Chemical and morphological analysis of PM revealed the presence of particles of biological origin, soot (Cr, Fe, Ni, Zn, Cd, Hg and Pb), salts (KCl) and soil resuspension (Al and Si). The integrated study methodology, employing environmental variables, such as PM and ryegrass, can be of help in the preparation of wide-ranging environmental diagnoses, in addition providing information needed to develop precautionary measures designed to minimize the effects of atmospheric pollution that takes into consideration the environment's supportive capacity and environmental quality.Keywords: air pollution, particulate matter, biomonitoring, lolium multiflorum, mev/eds. Estudos integrados empregando variáveis ambientais como o MP e o biomonitoramento com azevém podem auxiliar na elaboração de diagnósticos ambientais robustos, além de fornecer informações para o desenvolvimento de medidas de precaução que considerem a capacidade de suporte do meio ambiente e que visem à minimização dos efeitos prejudiciais da poluição atmosférica. Aquatic macroinvertebrate fauna is a relevant component of limnic continental aquatic ecosystems, playing an important role in several processes with relevant biocomplexity. The present study characterized the benthic macroinvertebrate fauna found in three hydric bodies in the Sinos river drainage basin regarding community structure. Sample was collected from January to December 2013 in three locations in the basin: the city of Caraá (29 °45'45.5"S/50°19'37.3"W), the city of Rolante (29°38'34.4"S/50°32'33.2"W) and the city of Igrejinha (29°36'10.84"S/50°48'49.3"W). Abiotic components (pH, dissolved oxygen and temperature) were registered and collected samples were identified up to family type. Average annual pH, dissolved oxygen and temperature were similar in all locations. A total of 26,170 samples were collected. Class Insecta (Arthropods) represented 85.5% of total sample. Platyhelmintes, Mollusca and Annelida samples were also registered. A total of 57 families were identified for the drainage basin and estimators (Chao-1, Chao-2 and jackknife 2) estimated richness varying from 60 to 72 families.Keywords: aquatic invertebrates, community structure, diversity indices. A comunidade de macroinvertebrados bentônicos da bacia hidrográfica do rio dos Sinos, Rio Grande do Sul, Brasil ResumoA fauna de macroinvertebrados aquáticos é um relevante componente dos ecossistemas aquáticos límnicos continentais, atuando em vários processos de importante biocomplexidade. O presente estudo caracterizou a fauna de macroinvertebrados bentônicos em três corpos hídricos da Bacia Hidrográfica do Rio dos Sinos, em relação a estrutura da comunidade. Amostragens foram realizadas de janeiro a dezembro de 2013 em três locais na bacia: Município de Caraá (29°45'45,5"S/50°19'37,3"W), Município de Rolante (29°38'34,4"S/50°32'33,2"W) e Município de Igrejinha (29°36'10,84"S/50°48'49,3"W). Variáveis abióticas (pH, oxigênio dissolvido e temperatura) foram registradas e os exemplares coletados identificados até família. As médias anuais de pH, oxigênio dissolvido e temperatura foram similares em todos os pontos. Um total de 26.170 exemplares foi amostrado. A Classe Insecta (Arthropoda) representou 85,5% do total. Platyhelmintes, Mollusca e Annelida também foram registrados. Foram determinadas 57 famílias para a bacia hidrográfica e os estimadores (Chao-1, Chao-2 e jackknife 2) projetaram uma riqueza variando de 60 a 72.Palavras-chave: invertebrados aquáticos, estrutura da comunidade, índices de diversidade.
