Anthracycline production by Streptomyces capoamus in batch fermentation

Abstract: Anthracyclines have been intensely studied worldwide due to their importance as anticancer agents. In this work investigation was made on the production of ciclamycin, an antitumour complex of the anthracycline class, produced by Streptomyces capoamus. The fermentation medium consisted of glucose, as the main carbon source, and soymilk, as the sole nitrogen source. Experiments were performed in a 5-liter batch bioreactor under nitrogen and carbon-limiting conditions. Increasing antibiotic activity was observed… Show more

“…IAA production by NKZ-259 was determined using nine different media types; ISP-1, ISP-2, ISP-4, glucose-yeast extract-malt extract (GYM) [39], Bennet’s medium, Gause’s No.1 medium [40], MS medium, Streptomyces medium [41] and tryptic soy medium [42]. NKZ-259 cells were inoculated in the above media at 28 °C on a rotary shaker with shaking at 200 rpm.…”

“…The strain Streptomyces capoamus is well known for the production of ciclamycin, ciclacidine, and capoamycin, an anthracyclin-class antitumor complex (Bieber et al, 1987;Gonclaves et al, 1968;Hayakawa et al, 1985;Martins and Maior, 2003) but to the best of our knowledge this strain has not been reported to produce antifungal compounds. The antifungal compound produced by this strain is fairly stable to the pH range 5-8.…”

“…Fructose, xylose, peptone, tryptone, and yeast extract have been reported earlier in the literature to enhance antifungal activity (Banna and Winkelmann, 1998;Vahidi et al, 2004). Unsuitable amino acids as nitrogen sources can inhibit good synthesis of secondary metabolites, whereas specific amino acids can in some cases enhance antibiotic production (Martins and Maior, 2003). Phenylalanine, isoleucine, methionine, and tyrosine are the most suitable amino acids for antifungal metabolite production whereas the addition of arginine, glycine, and valine repressed the synthesis of the antifungal metabolite (Table 2).…”

“…Anthracycline antibiotics were intensively studied because of their antitumor activity. Ciclamycine, one of the antitumor complex from the anthracycline class, is produced by Streptomyces capoamus [6]. A strain Streptomyces hydroscopcus 111-81 produces polyether complex, Hexaene and Nifimycine [7].…”
