Molecular characterization of MHC-DRB cDNA in water buffalo (Bubalus bubalis)

Abstract: In the present study, water buffalo MHC (Bubu)-DRB cDNA was cloned and characterized. The 1022 base long-amplified cDNA product encompassed a single open reading frame of 801 bases that coded for 266 amino acids. The Bubu-DRB sequence showed maximum homology with the BoLA-DRB3*0101 allele of cattle. A total of seven amino acid residues were found to be unique for the Bubu-DRB sequence. The majority of amino acid substitutions was observed in the β1 domain. Residues associated with important functions were most… Show more

“…in cattle ( Sharma et al, 2006; Huang et al, 2011 ). Maximum homology (99%) of 5′upstream region of buffalo TLR sequence with that of goat was observed to be in concordance with previous reports on TLR8 ( Dubey et al, 2012a,b ) and DRB * A ( Naskar et al, 2012 ) genes, wherein the water buffalo was observed to be phylogenetically more closer to goat than cattle. High homology of TLR8 region between riverine buffalo and goat suggests that TLR genes have evolved due to challenging by different types of species specific pathogens within populations of livestock species.…”

“…Under the DR region of the MHC complex, highly polymorphic β chain encoded by DRB genes is found (Zhao et al, 2011). DRA gene encodes α chain whereas β chain is coded by DRB gene (Naskar et al, 2012). Three BoLA (bovine leukocyte antigen) DRB genes (DRB1, 2 and 3) are reported in bovines but only DRB3 is functional (Singh et al, 2012).…”
