Natural infection of several Coffea species and hybrids and Psilanthus ebracteolatus by the coffee ringspot virus (CoRSV)

Abstract: Coffee ringspot is a minor coffee disease caused by the nuclear type of Brevipalpus mite-transmitted virus, Coffee ringspot virus (CoRSV). Recently outbreaks of the disease in some growing regions of the state of Minas Gerais, Brazil, were registered with qualitative and quantitative yield losses. Coffea arabica was the only species registered as natural host. A survey was made on a germplasm collection of Coffea and related species kept at the Centro de Café "Alcides Carvalho", Instituto Agronômico, Campinas,… Show more

“…To detect cytopathic effects caused by BTV-C and -N, lesioned tissues were processed, fixed in Karnovsky reagent, embedded in Spurr's epoxy resin and examined by TEM (Kitajima et al, 2001). Leaves were assayed by RT-PCR using specific primers for SvSRV (Ferreira et al, 2007), CoRSV (Kitajima et al, 2011) and ClCSV. Total RNA from independent plants inoculated with each virus was extracted using TRIZOL® Reagent (Thermo Scientific, WI, USA).…”

“…3 and 7). It is of particular interest whether a similar interaction between the N and P proteins can be observed for other candidate dichorhabdoviruses, including the citrus leprosis nuclear type, coffee ringspot, and Clerodendrum chlorotic spot viruses (54)(55)(56)(57).…”

“…Its genome is bipartite (5 and 9 kb) and of positive-sense single-stranded (ss)RNA. Two other BrTV-Ns, CoRSV (17) and CICSV (26), have been partially sequenced, exhibiting sequence similarities with OFV, and may belong to the same genus as other, as-yet-uncharacterized BrTV-Ns. Parts of the genomes of PFGSV (1) and SvRSV (11) have been sequenced, showing sequence similarities with that of CiLV-C. LigRSV and HGSV (23) have not been molecularly characterized yet but their cytopathology indicates that they are BrTV-C.…”
