Activity of cellulase from Thermoactinomycetes and Bacillus spp. isolated from Brassica waste compost

Abstract: Plant wastes present a high cellulose content, which is an ideal organic material for composting. Five strains of thermophiles from processed Brassica waste were isolated, and the hydrolytic activity on various cellulosic biomass substrata and their temperature profiles were determined. 16S rRNA sequencing identified these strains as Thermoactinomyces and Bacillus spp. Maximal cellulase activity corresponded to 2.3 U mL -1 of enzyme. The application of these strains on Brassica rapa residues demonstrates incre… Show more

“…Enzymatic hydrolysis of cellulose is a promising method for the conversion of waste cellulose to glucose. Many factors, such as the content of lignin, crystallinity of cellulose, degree of polymerization (DP), moisture content and particle size, affect the hydrolytic efficiency of the cellulose present in the lignocellulosic biomass (47). Lignin is one of the most abundant polymers in nature.…”

“…Several different pretreatment methods have been used to facilitate the subsequent enzymatic hydrolysis of lignocellulosic materials (11, 12). One of the most thoroughly investigated methods is alkali pretreatment in which the alkaline solution is used to remove lignin and various uronic acid substitutions on hemicelluloses, leading to the increased accessibility of enzymes to the hemicellulose and cellulose (4). Generally, alkaline pretreatment is more effective for the subsequent hydrolysis of agricultural residues and herbaceous crops than that for the wood materials (13).…”

“…Also Patagundi et al [14] isolated a total of 57 isolates from four different agricultural soil samples, three isolates dipicted maximum enzyme activity, they were identified as Bacillus cereus (0.440 U/ml), Bacillus subtilius (0.357 U/ml) and Bacillus thuringiensis (0.334U/ml). While Chang et al [15] isolated five isolates of thermophilic bacteria from cellulosic waste, and two of them (TA-1 and TB-2) were identified as Thermoactinomyces and Bacillus spp. Which showed maximum enzyme activity 2.07 and 2.3 U/ ml respectively.…”

“…Chang et al (2009) menemukan bahwa Bacillus sp yang diisolasi dari kompos limbah Brassica memiliki aktivitas selulase berkisar antara 1,9-2,33 U/ml. Demikian pula Pandey et al (2014) dan Ladeira et al (2015) Bacillus clausii adalah bakteri berbentuk batang, merupakan bakteri Gram posit if , m otil dan membentuk endospora (Hema & Shiny, 2012).…”
