Microsatellite markers for identification of Prunus spp. rootstocks

Abstract: Cultivar characterization for fruit trees certification requires fast, efficient and reliable techniques. Microsatellite markers (SSR) were used in the molecular characterization of 29 Prunus spp. rootstocks. The DNA from the rootstocks was analyzed using five pre-selected SSR primers (UDP96-005, UDP96-008, UDP96-013, UDP96-18 and UDP98-414) and revealed 81 alleles, which allowed each genotype to be identified. The UDP96-005 marker generated the most information, i.e., 23 well-distributed, polymorphic alleles … Show more

“…Thus, additional markers for identification may be useful and have been developed in several organisms. Most of them are specific DNA or RNA sequences obtained by different techniques (Li et al 2002;Randig et al 2004;Barik et al 2006;Vargas et al 2005;Bianchi et al 2004). Speciesmarker bands in native and SDS-PAGE gel electrophoresis of soluble proteins were described in fish by Hasnain et al (2005).…”

“…(Fang & Roose, 1997), grape (Lopez et al, 1999), cherry (Cantini et al, 2001;Struss et al, 2002), and pear (Monte-Corvo et al, 2001). Moreover, they have been used to study genetic diversity and relationships in apple (Hokanson et al, 1998), sweet cherry (Dirlewanger et al, 2002), Japanese apricot (Gao et al, 2004), plum (Ahmad et al, 2004b;Rohrer et al, 2004), almond (Xu et al, 2004), peach cultivars and rootstocks (Ahmad et al, 2004a;Bianchi et al, 2004;Marchese et al, 2005). They also have been efficient in testing the genetic origin of peach cultivars (Testolin et al, 2000) and the reconstruction of a grape pedigree (Sefc et al, 1998).…”
