Fish transport is one of the most stressful procedures in aquaculture facilities. The present work evaluated the stress response of matrinxã to transportation procedures, and the use of clove oil as an alternative to reduce the stress response to transport in matrinxã (Brycon cephalus). Clove oil solutions were tested in concentrations of 0, 1, 5 and 10 mg/L during matrinxã transportation in plastic bags, supplied with water and oxygen as the usual field procedures in Brazil. Clove oil reduced some of the physiological stress responses (plasma cortisol, glucose and ions) that we measured. The high energetic cost to matrinxã cope with the transport stress was clear by the decrease of liver glycogen after transport. Our results suggest that clove oil (5 mg/l) can mitigate the stress response in matrinxã subjected to transport. In order to determine the lethal dose (96-h LD 50 ) of the bacteria Aeromonas hydrophila to matrinxã, Brycon amazonicus, to be applied in challenge tests, 90 fish (63.23 ± 6.39 g) were divided into five treatments, with different bacterial solutions: T 1 -Control (0.9% NaCl saline solution); T 2 (4 x 10 11 cells/ mL); T 3 (5 x 10 11 cells/ mL); T 4 (1.36 x 10 12 cells/ mL) and T 5 (3.06 x 10 12 cells/ mL). Fish were previously anesthetized with benzocaine (60 mg L -1 ), inoculated in the peritoneal cavity with the bacterial suspensions and then distributed into fifteen 80-L test chambers, where the water variables were monitored and fish mortality was observed. The experiment was randomly designed in three replicates and the 96-h LD 50 was estimated according to the trimmed Spearman-Karber method. Water quality variables remained within adequate ranges for fish health and performance. Fish mortality rate increased with the bacterial concentrations of A. hydrophila (T 1 = 0%; T 2 = 16.66%; T 3 = 44.44%; T 4 = 72.22% and T 5 = 100%), and the first mortalities were observed after 57 h, although the signs of the bacterial infection were already observed 24 h after the inoculation. The results indicate that the 96-h LD 50 value of A. hydrophila to matrinxã is 6.66 x 10 11 cells/ mL. O delineamento experimental foi inteiramente casualisado com três réplicas e a DL 50 96-h foi estimada de acordo com o método Spearman-Karber. Durante o experimento os parâmetros físico-químicos da água permaneceram dentro das condições consideradas adequadas para o desenvolvimento e saúde dos organismos aquáticos. The objective was to evaluate the effects of different concentrations of ammonia (zero; 0.15; 0.30; 0.50; 1.00 mg L -1 NH3) on histology and morphology of gills and liver of juvenile Bryconops caudomaculatus. After 96 hours of exposure, the fishs were anesthetized and necropsied for collect gills and liver, which were embedded in paraffin. Morphological changes were observed on the secondary lamellae of the gills when concentrations greater than 0.15 mg L -1 , as hyperplasia, hypertrophy, epithelial displacement, lamellar fusion, swelling, lamellar aneurysm with rupture of the epithelium and increase in width of the lamellae were observed. The liver presented histopathologies (congestion in the capillaries and inflammatory processes) at the presence of 0.30 mg L -1 of NH3, compromising the vital functions, the metabolism, breathing processes, detoxification of the body, hematopoiesis and probably hindered osmoregulation. The results suggest that the changes in gill and liver can be used to monitor fish performance in intensive farming.Key words: morphological changes; contaminants; histology; toxicity. ANÁLISE HISTOLÓGICA DAS BRÂNQUIAS E DO FÍGADO DE Bryconops caudomaculatus EM DIFERENTES CONCENTRAÇÕES DE AMÔNIA RESUMOO objetivo foi avaliar os efeitos de diferentes concentrações de amônia não ionizada (zero; 0,15; 0,30; 0,50; 1,00 mg L -1 NH3) na histologia e morfologia das brânquias e fígado de juvenis Bryconops caudomaculatus. Após 96 horas de exposição, os peixes foram anestesiados, submetidos à necropsia, para coleta de brânquias e fígado, que foram incluídos em parafina. Alterações morfológicas nas lamelas secundárias das brânquias foram observadas em concentrações a partir de 0,15 mg L -1 , como hiperplasia, hipertrofia, deslocamento epitelial, fusão lamelar, edema, aneurisma lamelar com ruptura do epitélio e aumento na largura das lamelas. O fígado apresentou alterações histológicas (congestão nos capilares, processos inflamatórios e pigmentação endógena) na presença de 0,30 mg L -1 de NH3, e comprometeram as funções vitais, o metabolismo, processos de respiração, desintoxicação do organismo, hematopoese e provavelmente a osmorregulação. Os resultados sugerem que as alterações nas brânquias e fígado podem ser usadas para monitorar o desempenho de peixes em cultivo intensivo.
