ABSTRACT:This study aimed to evaluate the cytotoxicity of intracanal medications on L929 fibroblast cells at different periods of observation. The following experimental groups were studied: calcium hydroxide with camphorated paramonochlorophenol and glycerin (CPG); iodoform with glycerin (IG); calcium hydroxide with iodoform and distilled water (CIW); iodoform with distilled water (IW); calcium hydroxide with distilled water (CW); Otosporin ® (OT); and a control group composed of cells and culture medium. Eluates were prepared from each group and placed in contact with 1 x 10 5 cells/well for periods of 30 minutes, 12, 24, 48 and 72 hours, 5 and 7 days. After each experimental period, a cytotoxicity test was performed using methyltetrazolium (MTT) and a spectrophotometer at an optical density of 570 nm to analyze cell viability. The ANOVA and Tukey test with a significance level of 5% was used to analyze the data. At 30 minutes and at 12 hours, all groups were equal to the control group. At 24 hours, there was greater cytotoxicity in the IG group than in the control group (P<0.001). At 48 hours, only the OT group was cytotoxic (P <0.001). At 72 hours and at 5 days, the most cytotoxic groups were CW and OT. At 7 days, the IW and CPG groups were the least cytotoxic (P <0.001). With respect to experimental time, significant differences between 24 hours and 7 days were observed in all groups. Otosporin ® was the most cytotoxic medication, followed by calcium hydroxide with distilled water.

ResumoObjetivo: Avaliar o efeito da dentina sobre o pH e a atividade antimicrobiana de formulações com hidróxido de cálcio. Material e método: O pH e a atividade antimicrobiana das formulações (G1-polietilenoglicol, G2-glicerina e paramonoclorofenol canforado ou G3-clorexidina) foram analisados nas formulações puras (G1A, G2A e G3A) ou acrescidas de dentina em pó (G1B, G2B e G3B). Tubos de polietileno foram preenchidos com uma das formulações e imersos em água destilada. O pH foi analisado após 24 horas, 7, 14 e 21 dias. A atividade antimicrobiana foi avaliada sobre Enterococcus faecalis (ATCC 29212), através do teste de difusão em ágar. Os resultados foram submetidos aos testes de ANOVA e Tukey (p=0,05). Resultado: A dentina não exerceu efeito sobre o pH das formulações, exceto ocasionando a redução em G1, no período de 24 horas (p<0,05). Nas demais formulações, não houve interferência da dentina sobre o pH (p>0,05). Quando comparados entre os grupos, em 24 horas e 7 dias, G1A e G1B apresentaram menor pH (p<0,05). Em 14 dias, G2A, G2B, G3A e G3B demonstraram similar pH (p>0,05), assim como entre G3B e G1A (p>0,05). Em 21 dias, todas as formulações demonstram semelhante pH (p>0,05). Os resultados em ordem decrescente da atividade antimicrobiana foram G2A=G2B>G3A=G3B>G1A=G1B (p<0,05). Conclusão: A dentina não interferiu no pH e na atividade antimicrobiana das diferentes formulações com hidróxido de cálcio, exceto na associação com o polietilenoglicol, em que ocasionou a redução do pH apenas no período de 24 horas. A formulação com paramonoclorofenol canforado apresentou maior atividade antimicrobiana, seguida da clorexidina e do polietilenoglicol.Descritores: Cálcio; endodontia; Enterococcus faecalis; pH. Objective: To evaluate the antibacterial activity of four formulations of calcium hydroxide paste against microorganisms commonly found in infected root canals. Methods: To evaluate antibacterial activity through the agar diffusion method, pastes of calcium hydroxide were made from its pro-analysis form, diffused into four separate vehicles: distilled water, camphorated p-monochlorophenol, propylene glycol and Otosporin®, testing the antimicrobial activity of these on strains of Staphylococcus aureus, Bacillus subtilis and Enterococcus faecalis. After the incubation period, the presence or otherwise of inhibition zones were observed and their sizes in three stages: 24h, 48h and 72h. With this data, the median between the four dishes was obtained and the consequent value was submitted to Kruskal-Wallis nonparametric statistical analysis, with post-tests of Mann-Whitney and Bonferroni correction, at a significance level of 5%. Results: Only pastes with camphorated p-monochlorophenol and Otosporin vehicles caused the formation of significant inhibition zones, with medians of 8.0 mm. Against the strains of Enterococcus faecalis, only pastes with the camphorated p-monochlorophenol vehicle resulted in the formation of significant inhibition zones, with a median of 3.0 mm. Conclusion : Otosporin and CMCP vehicles provide greater antimicrobial potential to calcium hydroxide against the studied bacteria. However, only the Ca(OH)2 and CMCP combination was effective against all the strains, and can thus be regarded as the paste formulation with the greatest antimicrobial effectiveness in this study.
