A new galactose-specific lectin was purified from seeds of a Caesalpinoideae plant, Bauhinia variegata, by affinity chromatography on lactose-agarose. Protein extracts haemagglutinated rabbit and human erythrocytes (native and treated with proteolytic enzymes), showing preference for rabbit blood treated with papain and trypsin. Among various carbohydrates tested, the lectin was best inhibited by D-galactose and its derivatives, especially lactose. SDS-PAGE showed that the lectin, named BVL, has a pattern similar to other lectins isolated from the same genus, Bauhinia purpurea agglutinin (BPA). The molecular mass of BVL subunit is 32 871 Da, determined by MALDI-TOF spectrometry. DNA extracted from B.variegata young leaves and primers designed according to the B. purpurea lectin were used to generate specific fragments which were cloned and sequenced, revealing two distinct isoforms. The bvl gene sequence comprised an open reading frame of 876 base pairs which encodes a protein of 291 amino acids. The protein carried a putative signal peptide. The mature protein was predicted to have 263 amino acid residues and 28 963 Da in size.

RESUMODiferentes classes de proteínas são comuns em sementes de leguminosas, incluindo inibidores de tripsina e proteínas hemaglutinantes, as quais atuam sobre enzimas proteolíticas e sobre carboidratos da superfície celular, respectivamente. O objetivo deste trabalho foi quantificar, detectar e caracterizar parcialmente a ocorrência dessas proteínas em sementes de Tachigali plumbea, Sesbania exasperata e Ormosia costulata var. trifoliolata. Sementes das três espécies foram moídas e submetidas à extração salina (NaCl 0,15M -10 %, p/v). Os extratos totais obtidos foram utilizados para quantificar o conteúdo protéico, detectar a atividade residual da tripsina, a atividade hemaglutinante (AHE) e na obtenção do perfil protéico. A atividade residual da tripsina foi observada somente para T. plumbea e S. exasperata, cujos valores foram 4 e 19 %, respectivamente. A AHE foi detectada nos extratos das três espécies, sendo que os extratos totais de T. plumbea e S. exasperata, hemaglutinaram eritrócitos de rato, camundongo e hamster, enquanto que a espécie O. costulata hemaglutinou somente eritrócitos de hamster. O perfil protéico em SDS-PAGE revelou maior ocorrência de proteínas com massa molecular aparente de 10 a 30 kDa para T. plumbea e S. exasperata, enquanto que para O. costulata prevaleceram bandas protéicas com massa molecular variando entre 20-25 kDa. Conclui-se que os extratos totais de O. costulata e S. exasperata, pertencentes à subfamília Papilionoideae, apresentam menor conteúdo de inibidores de tripsina que T. plumbea (Caesalpinioideae) e, quanto à AHE, os resultados mostraram-se diferenciados, mesmo entre as espécies da mesma subfamília, tanto para a concentração mínima hemaglutinante quanto para a especificidade de interação com os eritrócitos.PalavRaS-chavE: Serinoproteinases, Sementes, Lectinas, Eritrócitos. RESUMOUm dos componentes da multimistura para suplementação alimentar de populações carentes é a farinha de folhas de mandioca (FFM) que possui elevado conteúdo em proteínas, vitaminas e minerais. Todavia, as folhas de mandioca também apresentam substâncias antinutritivas e/ou tóxicas, como cianeto, polifenóis, nitrato, ácido oxálico, hemaglutinina, saponinas e inibidores de tripsina. Objetivou-se neste trabalho extrair as proteínas da FFM, purificando-as em coluna cromatográfica e determinar sua atividade hemaglutinante e toxicidade. Foram testadas várias estratégias de extração e precipitação das proteínas, sendo que o maior teor protéico e atividade hemaglutinante foi obtido na extração com água destilada na proporção 1:20 (p/v) seguida da precipitação com sulfato de amônio a 80% de saturação. As proteínas precipitadas foram purificadas em coluna Q-Sepharose. Das quatro frações obtidas na purificação (I, II, III e IV), a I e a II apresentaram maiores atividades hemaglutinantes. As mesmas frações foram injetadas via intraperitoneal em camundongos com doses de 2 g (fração I), 3 g (fração II), 54 g (fração III) e 52 g (fração IV) para cada animal com 20g de peso médio, não sendo observadas mortes ou quaisquer efeitos adversos após 120h.Termos para indexação: Folha de mandioca, lectina, purificação, toxidez. ABSTRACTOne of the components of the multimixture to the feed supplementation of low-income populations is cassava leaf flour (FFM), with high content of proteins, vitamins and minerals. However, cassava leaves also present substance regarded as antinutritive and/or toxic, such as cyanide, polyphenols, nitrate, oxalic acid, hemagglutinin, saponins and trypsin inhibitors. The aim of this work was to extract proteins from FFM, purifying them in chromatographic column and determine their hemagglutinating activity and toxicity. A number of strategies of extraction and precipitation of proteins were tested; the highest protein content and hemagglutinating activity were obtained in the extraction with distilled water at the 1:20 ratio (p/v) followed by the precipitation with ammonium sulfate at 80% of saturation. The precipitated proteins were purified in Q-Sepharose Fast Flow column. Out of the four purification fractions (I, II, III e IV), the I and II activities presented higher specific activity. The same fractions were injected intraperitoneal via in mice of 20g weight with doses of 2 g (fraction I), 3 g (fraction II), 54 g (fraction III) and 52 g (fraction IV). No deaths or any adverse effects was observed after 120h.
