Does alfa lipoic acid prevent liver from methotrexate induced oxidative injury in rats?

Abstract: PURPOSE:To determine the antioxidant and anti-inflammatory effects of alfa lipoic acid (ALA) on the liver injury induced by methotrexate (MTX) in rats. METHODS:Thirty two rats were randomly assigned into four equal groups; control, ALA, MTX and MTX with ALA groups. Liver injury was performed with a single dose of MTX (20 mg/kg) to groups 3 and 4. The ALA was administered intraperitonealy for five days in groups 2 and 4. The other rats received saline injection. At the sixth day the rats decapitated, blood and … Show more

“…It has been shown that MTX showed renal and hepatic toxicity (Abdel‐Daim, Khalifa, Abushouk, Dkhil, & Al‐Quraishy, ). MTX is metabolised in liver and induced inflammation and liver damage and increased liver enzymes (Çakır et al., ). MTX metabolites have been confirmed to precipitate in kidney (Yuksel et al, ).…”

“…No information was found in the literature about the role of IL-1β and TNF-α in MTX oral mucositis. However, Çakir, et al 7 (2015) reported that MTX induced the amount of MDA and IL-1β and TNF-α gene expressions in the kidney tissue. In our experiment, HRE, which significantly prevented the increase of MDA, also significantly prevented the increase of IL-1β and TNF-α gene expressions.…”

“…The therapeutic effect of ALA has been the subject of research involving conditions such as atherosclerosis , joint diseases , AIDS , hepatic impairment , and diabetic polyneuropathy . Alpha‐lipoic acid has also been reported to be effective against conditions in which oxidative stress plays a role . It has also been reported that ALA might be a preventive agent in ulcerogenesis and inflammation‐associated tumor formation .…”
