Abstract: Purpose:To evaluate the effects of iloprost a prostacyclin analogue on the hepatic IR injury in rats. Methods: Forty male SpragueDawley rats (250-300 g) were divided into four groups each containing 10 rats;-(1) controls: data from unmanipulated animals; (2) sham group: rats subjected to the surgical procedure, except for liver I/R, and given saline; (3) I/R group: rats that underwent liver ischemia for 45 min followed by reperfusion for 45 min; (4) I-R/ Iloprost group: rats pretreated with iloprost (10 µg kg … Show more

“…Manasia et al 3 reported that low dose (0.075µg/ kg/min) iloprost increased the superior mesenteric blood flow in a systemic hypotension model cused by septic shock. Iloprost was also reported to have liver and kidney sparing properties in ischemia-reperfusion damage 4,5 . In patients with critical limb ischemia iloprost increased the endothelial progenitor cells in vivo and induced angiogenesis 6 .…”

“…Several studies have investigated the use of prostaglandins in LT. In particular, the prostaglandins E 1 and I 2 showed beneficial protective effects with respect to IRI by improvement of liver perfusion and protection of endothelial cells in vitro and in vivo (7–11). Both prostaglandins induce vasodilation, inhibit platelet aggregation and activation of leukocytes, and exert a variety of “cytoprotective” effects (12).…”

“…Prostacyclin also plays an important role in maintaining the proper microcirculation of the liver [33]. Gedik et al [12] reported that stimulation of prostacyclin expression by a prostacyclin analog iloprost alone was enough to protect the liver from IR. Furthermore, it has been reported that, antithrombin reduced liver IR injury by increasing the production of prostacyclin and prostaglandin E 2 through activation of COX-1 [19,26].…”
