The effect of zoledronate on bone remodeling during the healing process

Abstract: Purpose:To check the effect of zoledronate in bone remodeling during bone healing. Methods: Thirty rabbits were divided into two groups of fifteen animals each (control and experimental group respectively). Shaft osteotomy was performed on the cranial portion of the fibula of each animal. In the experimental group, a single dose of 0.04mg/kg of zoledronate was administered. In the control group, the same volume of bi-distilled water was administered. After one, two and four weeks, animals of both groups were k… Show more

“…Thus, we were not surprised to find that the expression of RANKL was significantly reduced in ONJ‐affected bone at both the protein ( P < 0.003) and mRNA ( P < 0.001) levels. Clinically, BP was also shown to enhance fracture repair of extracranial skeletal bone; this suggested that BP could differentially impact RANKL‐mediated bone remodelling in jaw and extracranial bone structures, although the RANK/RANKL‐OPG system regulates both CNC‐derived bone and MsC‐derived osteoblasts (Agarwala et al , 2002; Amanat et al , 2007; Matos et al , 2007). Experimentally, the application of zoledronic acid induced inflammation of the jaw bone, but did not affect the axial skeleton (Senel et al , 2010).…”

“…Early descriptions of ONJ as an avascular necrosis have been revised as a result of recent demonstrations of patent vascularization in ONJ (Hansen et al , 2006b). Indeed, amino‐BP treatment was found to improve vascularization of avascular bone in cases of femur head necrosis and to accelerate fracture repair in extracranial skeletal bone (Agarwala et al , 2002; Amanat et al , 2007; Matos et al , 2007). Several authors have reported an association between ONJ and local inflammation and infection; moreover, ubiquitous, intraoral actinomyces are commonly found in ONJ (Hansen et al , 2006a; Bisdas et al , 2008; Reid, 2009).…”

“…[7] showed increase in new bone formation when zoledronate was used in a distraction osteogenesis model and suggested that BS could have a positive effect for that drug. Zoledronate also improves the amount of bone in the metaphysis during bone healing, suggesting a positive effect on remodeling [8]. …”

“…Золедроновая кислота была исследована на различных моделях животных: на кроликах после остеотомии [24], при исследовании перестройки костных трансплантатов, а также после травматического повреждения у крыс без остеопороза и с моделированным остеопорозом [25,26].…”

“…Исследована регенерация у кроликов после проведения остеотомии малоберцовой кости и лечения однократной инъекцией золедроновой кислоты в дозе 0,04 мг/кг [24]. Контрольная группа животных получала дистиллированную воду.…”
