Effects of L-arginine oral supplements in pregnant spontaneously hypertensive rats

Abstract: Purpose:To evaluate the effects of L-arginine oral supplementation in spontaneously hypertensive pregnant rats (SHR). Methods: Thirty SHR and ten Wistar-EPM-1 virgin female rats were used in the study. Before randomization, females were caged with males of the same strain (3:1). Pregnancy was confirmed by sperm-positive vaginal smear (Day 0). Wistar-EPM-1 rats served as counterpart control (C-1). SHR rats were randomized in 4 groups (n=10): Group Control 2, non-treated rats; Group L-Arginine treated with L-arg… Show more

“…NOS substrate supplementation has beneficial effects on both blood pressure and renal disease in clinical trials (35,47,52) and in experimental models (12,14,27,32,40,42,50). In animals with ureteral obstruction, the administration of L-arginine increases GFR and renal blood flow to the postobstructed kidney and decreases the infiltration of macrophages of the renal parenchyma (29).…”

“…The l ‐arginine–NO pathway has been ascribed an important role in the development of hypertension and progressive renal disease, with NO synthase (NOS) substrate supplementation found to have beneficial effects in both clinical trials 58–60 and experimental models 61–66 . In a recent study, our group demonstrated that hypertensive animals with chronic PUUO or bilateral hydronephrosis had increased plasma levels of asymmetrical and symmetrical dimethylarginine (ADMA and SDMA, respectively) 67 .…”

“…This group (R+S) was also exposed to radiation on the eighth day of the experiment and was supplemented with L-arginine (Sigma-Aldrich®) from the beginning of the experiment (day 0) until death. L-arginne was administered by gavage in a dose of 0.65 g/Kg of body weight, diluted in distilled water [5][6][7][8] .…”
