Edema quantification by computerized morphometry as an evaluation parameter for the resistance of colon anastomoses

Abstract: PURPOSE: This work had the objective of verifying the validity of using computerized morphometry as a method of quantitative analysis of the interference of edema in scar formation following colon anastomosis. METHODS: Forty-five adult female Wistar rats were utilized, divided into three groups of 15 animals according to whether sacrifice was performed on the first, second or seventh postoperative day. Each group was subdivided into a main group consisting of 10 animals, and a control group consisting of five … Show more

“…Variables that can be quantitatively evaluated, such as colon crypt length, thicknesses of the different layers of the intestinal wall and number of caliciform cells can be determined objectively, thereby enabling conclusions that are more precise. Computer-assisted analysis has been used with various objectives and, in relation to conventional methods, it has the advantage of enabling fast and objective quantitative evaluation of the microscopic structures 22,23 . Few studies have evaluated inflammatory alterations in the colon wall of intestinal segments that have been taken out of function, by means of morphometric analysis methods 9,11,15 .…”

“…With the advent of computer-assisted image analysis software, it has become possible to objectively quantify the tissue expression of various proteins, stained using histochemical or immunohistochemical techniques 16,17 . With this method, it has become possible to quantify the expression of sulfomucins and sialomucins in the colon mucosa more precisely, thereby observing changes in total acid mucin content and the influence of the duration of exclusion, by comparing segments with and without fecal stream.…”

“…Little space is needed to house them; procreation occurs in large numbers and rapidly, ensuring almost identical lineages with animals that are genetically and physiologically similar. The management and maintenance costs are small compared with those for other animals; they are easy to obtain and handle, and are resistant animals 5 . In addition to these characteristics, it has been accepted since the study by Herrmann in 1964 6 that the fecal microflora of rats is very similar to that of humans.…”

“…Studies have habitually used animals of male gender [1][2][3][4][6][7][8][10][11][12][13]16,18 . even though there is no restriction on using females, purely from the point of view of location of the PP 5,14,21 .…”
