Purpose:To establish an inoculation model of Walker 256 carcinoma on cervix uteri and vagina of rats. Methods: Fifteen female rats were used, and assigned to three groups each one with fi ve rats: group A -rats with 4x10 6 cells of Walker 256 carcinoma without acid acetic inoculation; group B -rats with 2x10 6 cells of Walker 256 carcinoma with acid acetic inoculation and group C: rats with 4x10 6 cells of Walker 256 carcinoma with acid acetic inoculation. The day before tumor cells inoculation the rats from groups B and C were anaesthetized with diethylether and 0,3 ml of acetic acid was inoculated into their vaginas. Tumor cell inoculation into the vagina and cervix was done under general anesthesia with diethylether. Then a endocervical brush was used to scrape the vaginal wall and after that 0,3 ml of the liquid containing tumor cells was inoculated on the vagina and cervix. For the tumor analysis, animals were euthanized at day 12 following tumor cell implantation by an excessive inhalation of diethylether. Tumor was resected entirely and weighed and the tumors were then sectioned and counter stained with hematoxylin and eosin for histopathologic evaluation. It was also calculated the percentage of tumor equivalent to the body weight by the formula: P= tumor weight / body weight x 100. Data were analyzed by one-way analysis of variance -ANOVA. P values < 0.05 were taken to indicate statistical signifi cance. Results: Implantation and growth on GB and GC was 100% and on GA 20%. There was no statistical difference between GB and GC averages. Conclusion: According to the methods used, the Walker 256 carcinoma inoculation model into vagina and cervix have an implantation and growth rate of 100% when associated with previous acid acetic inoculation and there is no behavioral difference between using 2x10 6 or 4x10 6 cells on its inoculation. Métodos: Foram utilizadas 15 ratas fêmeas, virgens, adultas, pesando entre 200-250g, distribuídas em três grupos de estudo com cinco animais cada: grupo A (GA): ratas com tumor de Walker 256 em concentração de 4x10 6 sem ácido acético; grupo B (GB): ratas com tumor de Walker 256 em concentração de 2x10 6 células com ácido acético; grupo C (GC): ratas com tumor de Walker 256 em concentração de 4x10 6 células com ácido acético. No dia anterior à inoculação do tumor, foi realizada a inoculação de 0,3 ml de ácido acético a 10% na vagina das ratas de GB e GC; no dia seguinte, tanto estas como as ratas do grupo GA foram anestesiadas, feita a escarifi cação da parede vaginal com uma escova de endocérvice e inoculado 0,3ml de tumor na concentração de 4x10 6 células nos grupos GA e GC e 2x10 6 células no grupo GB. RESUMO ABSTRACTPurpose: An animal model to study bladder tumor with Walker 256. Methods: Bladders rats Wistar was catheterized via urethra and compression lesion of the wall bladder was realized with a little clamping after laparotomy. One total of the 0,3 ml suspension with 3 x 10 5 cels viable of the carcinossarcoma was instilled into each bladder. The animals were sacrificed after eight and 13 days. Results: The index of tumor incidence was 100% and the average of surviving was 14,5 days. Conclusion: The model estudied was efficient and will can to take subsidy the study experiemental in treatment of local invasise bladder cancer.

Purpose:To elaborate an experimental model of pulmonary carcinogenesis in Wistar rats. Methods: Male Rattus norvegicus albinus, Wistar lineage was carried through an intra-pulmonary instillation of the Benzo[a]pyrene (B[a]P) dilution in alcohol 70%, a polycyclic aromatic hydrocarbon widely known by its power of tumoral induction. Three experimental groups had been formed with 08 animals each: Control Group (Alcohol 70%); B[a]P Group 10 mg/kg; e B[a]P Group 20mg/ kg, submitted to euthanasia 08, 10, 12 and 14 weeks after the experimental procedure. The pulmonary sections had been colored by hematoxilin-eosin (HE) and submitted to the morphometrical analysis to describe the tissue alterations. Results: The presence of diffuse inflammatory alterations was observed in all groups, however, at the analysis of the pulmonary tissue of the experimental groups, it had been observed hyperplasic alterations (BALT hyperplasia), and in one of the animals of the experimental group 20mg/kg (12 weeks), it was noticed the presence of cellular epithelial tracheal pleomorphism, suggesting the adenocarcinoma formation in situ. Conclusion: The main secondary alterations to the intra-pulmonary instillation of B[a]P in Wistar rats were: cellular proliferation, inflammatory alterations of several degrees and nodular lymphoid hyperplasias. The association of an activator agent of the pulmonary metabolic reply is necessary to establish the ideal reply-dose to the development of the lung cancer. Key words: Carcinogenesis. Lung Cancer. Métodos: Rattus norvegicus albinus, linhagem Wistar foram submetidos a instilação intra-pulmonar da diluição em álcool 70% de Benzo[a]pireno (B[a]P), um hidrocarboneto aromático policíclico amplamente conhecido por seu poder de indução tumoral. Foram formados três grupos experimentais com 08 animais cada: Grupo Controle (álcool 70%); Grupo B[a]P 10 mg/kg; e Grupo B[a]P 20mg/ kg, submetidos a eutanásia 08, 10, 12 e 14 semanas após o procedimento experimental. As secções pulmonares foram coradas por HE e submetidas a análise morfométrica para descrição das alterações teciduais. Resultados: em todos os grupos observou-se a presença de alterações inflamatórias difusas, porém na análise do tecido pulmonar dos grupos experimentais, observou-se alterações hiperplásicas (hiperplasia de BALT), e em um dos animais do grupo experimental 20mg/kg (12 semanas) notou-se a presença de pleomorfismo celular epitelial traqueal, sugerindo a formação de adenocarcinoma in situ. Conclusão: as principais alterações secundárias à instilação intra-pulmonar de B[a]P em ratos Wistar foram: proliferação celular, alterações inflamatórias de diversos graus e hiperplasias nodulares linfóides. A associação de um agente ativador da resposta metabólica pulmonar pode ser necessária para estabelecimento da dose-resposta ideal ao desenvolvimento do câncer de pulmão. Descritores: Carcinogênese.
