Abstract:Fucan is a term used to denominate a family of sulfated polysaccharides rich in L-fucose. The brown alga Spatoglossum schröederi, Dictyotaceae, synthesizes three heterofucans named A, B, and C. Fucan A is a non-anticoagulant heterofucan which possesses potent antithrombotic (in vivo) and antiproliferative (in vitro) activities. However, its toxicity in vivo has not been determined. The present study examined the acute and subchronic toxicity of the fucan A in Wistar rats after subcutaneous administration. After that, the animals were killed and examined. The results showed in the acute study that fucan A did not cause general adverse effects and mortality in the concentrations 0, 20, 100, 1000, and 2000 µg/g body weight per rat for seven days. Regarding the subchronic study, the data showed that the fucan A did not cause any change in hematological and biochemistry parameters, as well as in the morphology, and in the size of the rat's organs analyzed at a concentration of 20 µg/g body weight per rat during a 62-day period. In conclusion, this study indicates this heterofucan is a compound with potential pharmacological value that has no toxicity in vivo.

RESUMO -Frutos não tradicionais como os deCereus jamacaru DC são fontes de fitonutrientes importantes para a saúde, além de constituir uma alternativa alimentícia em épocas secas até para os animais. Este trabalho teve por objetivo avaliar a qualidade nutricional e funcional de frutos de mandacaru em diferentes estádios de maturação (Verde Claro (VC), Inicio de Pigmentação (IP), Inicio de Pigmentação Vermelho (IPV), Pigmentado Vermelho (PV), Totalmente Vermelho (TV)) , visando agregação de valor através da diversificação de seus usos na região e explorando o potencial dos frutos como alimento funcional. Os frutos de mandacaru foram colhidos no município de Campina Grande-PB e Barra de Santa Rosa-PB. O delineamento experimental utilizado foi inteiramente casualizado e doze frutos (quatro repetições de três frutos) foram colhidas de cada estádio de maturação. Os dados obtidos das variáveis físicas, físico-químicas, polifenóis extraíveis totais e atividade antioxidante foram submetidos à análise de variância pelo teste F e as médias foram comparadas pelo teste de Tukey em até 5% de probabilidade. Os frutos de mandacaru apresentam elevado rendimento de polpa no estádio de maturação maduro (TV-totalmente vermelho) e coloração de casca que evoluiu de verde pigmentado para a coloração avermelhada com o avanço da maturação. Os sólidos solúveis, a relação SS/AT e pH dos frutos aumentou com o avanço da maturação, enquanto a acidez titulável diminuiu, todos estes tendendo a se estabilizar no estádio IPV, indicando ser este o ponto de colheita. Isso torna esses frutos adequados para o consumo fresco e para serem processados. Os frutos de mandacaru apresentam um alto conteúdo de polifenóis extraíveis totais em todos os estádios de maturação e uma elevada atividade antioxidante total, justificando assim o seu consumo, além de servir como estímulo ao seu melhor aproveitamento. ResumoO objetivo deste trabalho foi verificar em sangue de rato a compatibilidade entre a metodologia automatizada e a não automatizada. Foram realizadas contagens das variáveis hematológicas de 10 ratos Wistar, Rattus norvegicus. Para o método automatizado foram utilizados os aparelhos ABX-Pentra-60c+ e Celldyn-3200. Para o método não automatizado foi utilizado o hemocitômetro nas contagens de eritrócitos, de leucócitos e de plaquetas, utilizando o líquido de Hayem (1:200), o líquido de Türk (1:20) e o líquido de Brecher e Cronkite (1:200), respectivamente. A determinação do hematócrito foi obtida utilizando-se uma centrífuga de micro-hematócrito, a concentração de hemoglobina pelo método de cianometa-hemoglobina (líquido de Drabkin) e os índices hematimétricos absolutos calculados através dos resultados da série vermelha. A contagem diferencial de leucócitos foi realizada em extensões de sangue (panótico rápido). Os dados foram analisados através da variância -ANOVA, seguida pelo teste Tukey, com nível de significância de 5%. Os valores obtidos pelo hemocitômetro comparadas aos contadores hematológicos mostraram diferenças significativas (p < 0,05) para neutrófilos, eosinófilos e basófilos. Os resultados permitiram concluir que as contagens realizadas com os aparelhos utilizados foram confiáveis e rápidas, mas não são indicadas para a contagem diferencial de leucócito.Palavras-chave: automação, hematologia, hemograma, sangue. AbstractThe aim of this study was to verify blood compatibility between automated and non-automated methodology in rats. Counts of haematological variables of 10 Wistar rat, Rattus norvegicus were made. For the automated method were used the ABXPentra-60c + equipment and Celldyn-3200. For non-automated method the hemocytometer was used to count the erythrocytes, leukocytes and platelets using the Hayem liquid (1: 200), the Türk liquid (1:20) and the Brecher and Cronkite liquid (1: 200), respectively. The determination of the hematocrit was obtained by using a microhematocrit centrifuge, hemoglobin concentration by cyanmethaemoglobin method (Drabkin liquid) and absolute RBC indices calculated by the results of red series. The differential leukocyte count was performed on blood extensions (fast Panotic). The data was analyzed through variance -ANOVA and then by Tukey test at 5% significance level. The values obtained by hemocytometer compared to hematological counters revealed significant differences (p < 0.05) for neutrophils, eosinophils and basophils. Through the results obtained, it was concluded that the counts made with the devices used were reliable and fast, but are not indicated for the differential count of white blood cell.
