RESUMO:Este estudo descreve a anatomia e analisa a histoquímica dos órgãos vegetativos de Priva lappulacea (L.) Pers. Secções transversais e paradérmicas, à mão livre, de material fresco e fixado foram utilizadas para as análises anatômicas e histoquímicas, usando microscopia óptica. Os caracteres anatômicos são comuns àqueles descritos para a família, podendo ser utilizados como diagnóstico na sua identificação. Destacam-se, nas folhas, os tricomas, glandular e não-glandular, estômatos anomocíticos, em ambas as faces da epiderme, e mesofilo dorsiventral. As expansões no pecíolo são acrescentadas à diagnose da espécie. Estudo fitoquímico realizado com as partes aéreas mostrou a presença de triterpeno (ácido ursólico) e esteróide (β-sitosterol), iridóides (ipolamida e catalpol), açúcar redutor (glucose), flavonóide (luteolina) e fenilpropanóide (verbascosídeo). Nas raízes foi encontrada apenas glucose e dois iridóides estiveram presentes no caule e folhas. Alcalóides, saponinas, cumarinas, derivados cinâmicos, proantocianidinas condensadas e leucoantocianidinas não foram constatados nas partes analisadas. A descrição anatômica e os testes histoquímicos são inéditos para P. lappulacea. Unitermos:Priva lappulacea, Verbenaceae, morfoanatomia, perfil fitoquímico.ABSTRACT: "Morphoanatomy, histochemistry and phytochemical screening of Priva lappulacea (L.) Pers. (Verbenaceae)". This study describes the anatomy and analyzes the histochemistry of the vegetative organs of Priva lappulacea (L.) Pers. Freehanded transversal and paradermal sections of fresh and fixed material were used for the anatomical and histochemical analyses, using optical microscopy. The anatomical characters are common to those described for the family, being able to be used to diagnosis in its identification. The leaves are highlighted by the presence of glandular and non-glandular trichomes, anomocytic stomata on both faces of the epidermis and dorsiventral mesophyll. The expansions in the petiole are added to the diagnosis of this species. The phytochemical study carried out with aerial organs showed the presence of triterpene (ursolic acid) and steroid (β-sitosterol), iridoids (ipolamide and catalpol), reducing sugar (glucose), flavonoid (luteolin) and fenilpropanoid (verbascosides). In the roots only glucose was found and two iridoids are present in stem and leaves. Alkaloids, saponins, cumarines, cinamic acid derivates, condensed proantocianidines and leucoantocianidines were not checked in the analyzed parts. The anatomical description and the histochemical tests are unpublished for P. lappulacea.

Background: Practitioners of syncretic religions often incorporate plants into their sacred rituals, a unique practice that helps keep them connected to their cultural history. An important example of ritual plant use is found in the botánicas (religious stores that cater to practitioners of Afro-Caribbean diaspora religions) of Miami-Dade County, Florida (USA).Methods: We examined ritual plants sold in a sample of six botánicas in Miami-Dade County from November 2018 until April 2019. We addressed the following questions: 1) What is the ratio of native to non-native plant species found in the botánica stores; 2) What is the ratio of invasive exotic to non-invasive exotic plant species found in the botánica stores; and 3) What is the geographical origin of plant species found in the botánica stores?Results: Given the study's location in South Florida, the frequency of local native flora sold as ritual plants was significantly smaller than initially predicted (27% vs. 65%). Likewise, actual frequency of invasive species was much higher than the expected frequency (32% vs. 19%). We also discovered that most of the taxa collected during the study were Neotropical (50%) and Asian (25%) in origin, while very few African taxa were documented (7 out of the 93 species identified). Conclusion: These botánicas highlight the reliance of Afro-Caribbean syncretic religions in Miami-Dade County on locally common plants, particularly those taxa from the Neotropics. Future studies should further investigate how these plants are being cultivated, traded, and used by the informal economy associated with urban botánicas.

Arginase is a metalloenzyme that plays a central role in Leishmania infection. Previously, rosmarinic and caffeic acids were described as antileishmanial and as a Leishmania amazonensis arginase inhibitor and now, we describe the inhibition of arginase in L. amazonensis by rosmarinic acid analogs (1-7) and new caffeic acids derived amides (8-10). Caffeic acid esters and amides were produced by means of the engineered synthesis in E. coli and tested against L. amazonensis arginase. New amides (8-10) were biosynthesized in E. coli cultured with 2 mM of different combinations of feeding substrates. The most potent arginase inhibitors showed Ki(s) between 2 - 5.7 μM. Compounds 2-4 and 7 inhibited L-ARG through a noncompetitive mechanism, and 9 showed a competitive inhibition. By applying an in silico protocol we determined the binding mode of compound 9. The competitive inhibitor of L-ARG targets key residues within the binding site of the enzyme establishing a metal coordination bond with the metal ions as well as a series of hydrophobic and polar contacts supporting its micromolar inhibition of L-ARG. These results highlight that the dihydroxycinnamic-derived compounds can be used as the basis for the development of new drugs using a powerful tool based on the biosynthesis of arginase inhibitors.
