Antimicrobial Activity of Copaifera Spp. Against Bacteria Isolated From Milk of Cows With Mastitis

The antimicrobial activities of the oleoresin (OR) and the essential oil (EO) of Copaifera spp. were checked against microorganisms isolated from milk samples of cows diagnosed with grade III subclinical mastitis. The OR had good antimicrobial activity (MIC ≤ 100 µg/mL) against samples of coagulase-positive Staphylococcus, coagulase-negative Staphylococcus, Streptococcus of groups C, F, and G, and Corynebacterium spp. Meanwhile, the EO had good antimicrobial activity (MIC≤100 µg/mL) against coagulase-negative Staphylococcus and Corynebacterium spp. The OR and the EO were inactive and weak to inactive, respectively, against Escherichia coli. Overall, the OR had better antimicrobial activity than the essential oil against the 55 bacterial isolates (p<0.0001). The GC/MS analysis identified sesquiterpenes in EO and by the ESI FT-ICR MS method, the identification of diterpenic acids in OR was possible. Therefore, this raw plant material is promising for the development of phytotherapeutic drugs against bovine mastitis.Keywords: bovine mastites; ESI FT-ICR MS; essential oil; minimum inhibitory concentration; oleoresin. A atividade antimicrobiana da oleorresina e do óleo essencial de Copaifera spp. foi verificada frente aos micro-organismos isolados de amostras de leite de vacas diagnosticadas com mastite subclínica grau III. A oleorresina de Copaifera ssp. apresentou boa atividade antimicrobiana (CIM≤100 µg/mL) frente às amostras de Staphylococcus coagulase positivo, Staphylococcus coagulase negativo, Streptococcus do grupo C, F, G e Corynebacterium spp. Já o óleo essencial de Copaifera spp. apresentou boa atividade antimicrobiana (CIM≤100 µg/mL) frente aos Staphylococcus coagulase negativo e Corynebacterium spp. Em relação à Escherichia coli, a atividade antimicrobiana da oleorresina e o óleo essencial foi inativa e fraca à inativa, respectivamente. Conclui-se que, frente às 55 bactérias isoladas, a olerresina apresentou melhor atividade antimicrobiana do que o óleo essencial de Copaifera spp. (p<0,0001). A análise por CG/EM identificou sesquiterpenos no óleo essencial e pelo método ESI FT-ICR MS, foi possível identificar ácidos diterpenos na oleorresina de Copaifera spp. Por fim, sugere-se que a oleorresina de Copaifera spp. é uma matéria-prima vegetal promissora no desenvolvimento de medicamento fitoterápico para o tratamento de mastite bovina.Palavras-chave: concentração inibitória mínima; ESI FT-ICR MS; mastite bovina; óleo essencial; oleorresina.

The Copaiba oil-resin (Copaifera sp.) is widely used in folk medicine, especially as a healing, antiinflammatory, antiseptic, to treat ulcers and skin diseases. Increased gastric emptying was observed with oilresin of Copaifera langsdorfii, highlighting those others species from the genus of Copaifera can act over the gastrointestinal tract. This study evaluated the activity of the Copaifera sp oil-resin on gastric emptying in Rattus novergicus. Eighteen male rats were fasted for 24h and orally received Tween-80 and Copaiba oil-resin 100 and 200 mg/kg; one hour later they all received phenol red (PR) 0.5 mg/mL. Then, the parts of gastrointestinal tract (stomach and intestine) were analyzed. The results showed that Copaifera sp oil-resin at 200 mg/kg increased the gastric emptying in Rattus novergicus.

O objetivo deste estudo foi avaliar o efeito antimicrobiano de dois diferentes óleos de copaíba sobre isolados de Escherichia coli obtidos de leite mastítico. O ensaio antimicrobiano foi realizado utilizando o teste de difusão em agar, testando três soluções: um controle negativo e duas soluções dos diferentes óleos de copaíba. O teste foi realizado com 27 isolados de E. coli. Oito isolados (29.63%) foram inibidos pela solução com óleo de C. langsdorffii e sete pela solução com óleo de C. officinalis. Os resultados do presente estudo sugerem o óleo de copaiba como uma potencial fonte de novos e seletivos agentes para o tratamento da mastite, após posteriores testes em modelos in vivo. An antimicrobial assay was performed using an agar diffusion test, testing three solutions: a negative control and two solutions of different copaiba oils. This test was performed with 27 mastitic E. coli isolates. Eight isolates (29.63%) were inhibited by C. langsdorffii oil solution and seven by C. officinalis oil. The results of the present in vitro study suggest that copaiba oils may be potential sources of new and selective agents for the treatment of mastitis, pending further testing in in vivo models.
