Effect of concentration and exposure period to butyrolactone I on meiosis progression in bovine oocytes

Abstract: The effect of concentration and exposure period of bovine oocytes to butyrolactone I (BLI) on meiotic block and in vitro maturation (IVM) kinetics was studied. In experiment 1, all oocytes were at germinal vesicle stage (GV), after 6h in culture with 0, 50 and 100µM BLI. After 12h, all oocytes cultured with 50 and 100µM BLI remained in GV. After 24h, less oocytes were in GV with 50µM (82%) than with 100µM BLI (99%, P<0.05). In experiment 2, after 6h IVM, 93% of control oocytes (IVM only) were in GV, while trea… Show more

“…2011), and after release of meiosis inhibition, it is possible that some mechanisms under control of these kinases are disturbed. Similar to accelerated meiosis resumption reported for such treatments (Adona and Leal 2006), degradation of certain transcripts could be also accelerated.…”

“…Previous studies have shown that meiosis resumption is accelerated as GVBD is observed earlier during maturation (Hashimoto et al. 2002; Adona and Leal 2006; Adona et al. 2008a).…”

“…Temporarily blocking meiosis in oocytes has been shown to accelerate the kinetics of the cell cycle in these cells irrespective of the duration of blocking culture or the kind of drugs used (Mayes and Sirard 2001; Adona and Leal 2006), causing a shortening of the duration of maturation with no effect on development. This phenomenon was well established by observations from our laboratory (Adona and Leal 2006; Adona et al. 2008a; b) and others (Ponderato et al.…”

“…SD, standard deviation. inhibitory substances has been tested, however no advances have been achieved in embryo production (Adona & Leal, 2006;Gharibi et al, 2013;Rose et al, 2013;Ulloa et al, 2014). Unlike other inhibitors, cilostamide, which inhibits PDE-3A, specifically acts in the oocyte to inhibit cAMP degradation and therefore retains meiotic progression (Sasseville et al, 2009).…”
