The aim of this work is to study the effect of explant age and exogenously applied IBA on the growth and rooting potential of potato apical stem cuttings for production of first- generation seed potatoes. The experimental protocol consists in taking apical stem cuttings from potato plantlets at different physiological ages (30 and 60 days old) in interaction with the growth regulator indole-3-butyric acid (IBA) at different concentrations (100, 200, and 300 ppm) to study the production of potato minitubers. The results of the present study showed that mini-cuttings from young potato plants (30 days old) have a higher rooting capacity than mini-cuttings from mature plants. In addition, IBA concentrations, especially at 300 ppm, have a significant effect on most of the characteristics studied, e.g. rooting %, root number, root length, shoot length, and number of leaves. Regarding yield attributes, application of IBA at 300ppm gave the highest average tuber number (6.66) and tuber yield (142.97g) was higher in 30days old explant cuttings. The data presented in this study may encourage potato growers to use potato stem cuttings as a new, cheaper method of growing potatoes instead of expensive imported seed tubers. It is suggested that the use of apical stem cuttings can be a quick and effective method for mass propagation and to increase the yield of minitubers in the seed potato production cycle.

The production of mini-tubers under soilless cultivation system increased the availability of pathogen-free seed potatoes. The objective of this study was to evaluate the rooting capability of mini-cuttings of potato clones as a function of the irrigation solution and the physiological age of the mother plant, under soilless cultivation system. Potato mini-cuttings were collected from young and mature plants of the cultivars Asterix and Macaca and the advanced clone SMINIA 793101-3. The young plants were newly acclimatized individuals and the mature plants were at the stage of producing mini-tubers. In the first experiment, mini-cuttings originating from summer pruning of mature plants were grown under closed soilless cultivation system irrigated with a nutrient solution developed for the production of potato mini-tubers and a solution comprised of tap water only. The experiment was conducted in a 3 × 2 factorial arrangement (clones and irrigation solutions) in the complete random design with four replicates of 15 mini-cuttings. In the second experiment, the young and mature mini-cuttings were established under a closed soilless cultivation system irrigated with nutrient solution. The experiment was conducted in a 3 × 2 factorial arrangement (clones and physiological age of the mother plant) in the complete random design with four replicates of 15 mini-cuttings. In both experiments, the percentage of rooting and survival of mini-cuttings, number of roots and length of the longest root were assessed at 21 days of cultivation. Potato mini-cuttings can be rooted under closed soilless cultivation system irrigated with nutrient solution or tap water. Mini-cuttings of young plants have higher rooting capability compared to the ones taken from mature plants. The loss of rooting capability depends upon the potato cultivar.

Potato is the world's most important non-cereal food crop, and therefore, it is considered one of the major food sources for humankind. Its conventional propagation is asexual, by using the tuber, which allows the accumulation and dissemination of pathogens to new cultivation areas. This fact not only impairs the yield of this solanaceous plant, but also threatens the maintenance of genotypes for commercial or breeding purposes. Due to the impossibility of using botanical seed, conservation and exchange of germplasm of this species by means of conventional methods are not feasible. In all potato-producing regions, the demand for high-quality tubers has been paramount to ensure crops production. Thus, biotechnological techniques based on tissue culture are very important. Plant tissue culture offers alternative methods of propagation by in vitro techniques that provide production and multiplication of material with high sanity. Thus, this literature review summarizes the history and current situation of tissue culture techniques applied to potato crop. Besides clonal multiplication, this biotechnological tool makes available initial indexed material to breeding programs and certified seed potato, and facilitates the exchange and conservation of germplasm. For all these reasons, the use of these techniques in potato production chain directly benefits producers by providing high-quality propagules.
