Cardiac and vascular effects of diltiazem, dobutamine and amrinone, drugs used after myocardial revascularization

Abstract: Hemodynamic care during postoperative management of myocardial revascularization should include vasorelaxing drugs to insure adequate graft and coronary flow, and stimulation of stroke volume to maintain vascular perfusion pressure. We tested the cardiac (inotropic and lusitropic) and vascular (relaxant) effects of diltiazem (0.1 nM to 0.1 mM), dobutamine (10 µM to 10 mM) and amrinone (10 µM to 1 mM) on isolated rat atria and thoracic aorta, and also on isolated human saphenous vein (HSV) and human mammary art… Show more

“…Diltiazem (DTZ) is a non-dihydropyridine, calcium channel blocker, used in the treatment of hypertension, angina pectoris, and some types of arrhythmia [17]. The pharmacological profile of DTZ includes interference of calcium channels and blockage of influx of intracellular calcium: essential for smooth muscle contraction.…”

“…In our preliminary studies iloprost caused a 5.57% vasoconstriction at 10 )5 M of concentration and papaverine did not respond until the concentration 10 )5 (data not shown). After the contractile response had reached plateau, each ring was randomly allocated to treatment with cumulative concentrations of iloprost (10 )9 -10 )6 M, n ¼ 10) papaverine (10 )5 -10 )4 M; n ¼ 10), diltiazem (10 )9 -10 )4 M, n ¼ 11) [14] or ethanol alone (equal concentration in iloprost solution, n ¼ 10), a solvent for iloprost. The relaxation was expressed as percentage reversal of the KCl contraction.…”
