Abstract: To determine if radiocontrast impairs vascular relaxation of the renal artery, segments (4-5 mm in length) of canine renal artery were suspended in vitro in organ chambers to measure isometric force (95% O 2 /5% CO 2 , at 37ºC). Arterial segments with and without endothelium were placed at the optimal point of their length-tension relation and incubated with 10 µM indomethacin to prevent synthesis of endogenous prostanoids. The presence of nonionic radiocontrast (iohexol, Omnipaque 350, 1 ml in 25 ml control s… Show more

“…Alterations of microvasculature in the renal glomerulus are thought to be involved in renal hypertension; however, an association between the ILA and hypertensive atherosclerosis and hypertension has also been reported ( Jensen et al ., 1994 ). Responses in the ILA have been studied previously in dogs and rabbits ( Caffrey et al ., 1990 ; Discigil et al ., 2004 ), but not in mice. To measure force development in the ILA, the sensitivity of the force transducer was increased as the tissue‐mounting wires were minimized (30 μm in diameter, 1.0–1.3 mm in length).…”

“…Thus, it was necessary to develop force measurement techniques that can be applied to isolated intrarenal microvessels. Force measurement in intrarenal arteries has previously been documented for both the dog ( Discigil et al ., 2004 ) and the rabbit ( Caffrey et al ., 1990 ). In these investigations, the interlobar artery (ILA) was used as a typical intrarenal artery.…”

“…The use of contrast agent can lead to acute kidney dysfunction [ 66 ]. Ionic contrast agents could impairs renal artery relaxation by interfering with both cAMP-mediated and cGMP-mediated pathways [ 67 ].…”

“…Each aortic segment was cut into approximately 3 mmwide rings. These rings were suspended in the carbonated (95% oxygen, 5% carbon dioxide) Krebs solution (118.3 mM NaCl, 4.7 mM KCl, 1.2 mM MgSO4, 1.22 mM KH2PO4, 2.5 mM CaCl2, 25.0 mM NaHCO3 and 11.1 mM glucose, Sigma-Aldrich), which was heated and circulated at 37 °C (Heater and Circulatory system -May WBC 3044V3), without delay (18). Each aorta ring was passed through two stainless steel hooks in a 25 ml organ bath filled with Krebs solution (May 99 IOBS Ankara, Turkey), and suspended to the organ bath.…”
