Role of gelatinases MMP-2 and MMP-9 in tissue remodeling following acute lung injury

Abstract: Acute lung injury is characterized by a severe disruption of alveolocapillary structures and includes a variety of changes in lung cell populations. Evidence suggests the occurrence of rupture of the basement membranes and interstitial matrix remodeling during acute lung injury. The dynamic equilibrium of the extracellular matrix (ECM) under physiological conditions is a consequence of the balance between the regulation of synthesis and degradation of ECM components. Matrix metalloproteinases (MMPs) represent … Show more

“…It degrades type IV collagen, proteoglycan, and laminin. MMP-9 is produced by tumoral cells as well as inflammatory cells such as neutrophil, eosinophil, monocyte, lymphocyte and alveolar macrophage (Chintala et al 1999;Corbel et al 2000;Apakkan Aksun et al 2001;Bayramoglu et al 2009Bayramoglu et al , 2011.…”

“…MMP-2 and MMP-9 are known to degrade type IV collagen and gelatin, which are major components of the extracellular matrix (53)(54)(55). Their excess activity has been thought to contribute to the development of emphysema through extensive destruction of the extracellular matrix and remodeling of lung parenchyma (54,56,57). Therefore, we propose that the alveolar enlargement in Ig-Hepta Ϫ/Ϫ may result from the overactivation of the MMPs, which causes alveolar tissue destruction and remodeling.…”

“…Knockout mice showed significant reductions in the expression of macrophage inflammatory protein-1 alpha (MIP-1α), MIP-1β, and MCP [86]. Additionally, neutrophil and macrophages are known to activate the MMP-2 and -9 secretion [87,88]. However, addition of inflammatory cytokines to cells upregulates the expression of MMP-2 and -9 along with other various cytokines and chemokines, and administration of IL-1β to diabetic animal upregulates the expression of MMP-2 and -9.…”
