Show more

“…This medium also contained boric acid and calcium nitrate in concentrations commonly used in the specific literature [17][18][19][20][21][22] Pollen germination test was performed on slides in a wet chamber maintained in Petri dishes, which were left at room temperature (25˚C -27˚C) for 12 h and up to 24 h. Pollen grains were placed to germinate in treatments tested (see below), and after respective incubation times were counted pollen grains which have germinated on a randomly sample of 500 grains in three slides by each treatment. For the purpose of counting, germinated pollen was considered when the pollen tube length was equal to or more than twice the diameter of the each pollen grain.…”

“…No viability or artificial in vitro germination test is quite satisfactory, especially after pollen has been stored, for the following reasons: the chemical tests employ dyes that react with chemical constituents or structures whose presence may not reflect the ability of the pollen to germinate; samples of PG that germinate in vitro may not produce sufficient elongation of the pollen tube as to affect fertility (Franzon & Raseira 2006). On the other hand, for some species pollen samples that appear to be non-viable when tested in vitro can produce large percentage of seeds, and the pollen stored can differently germinate in repeated samples or in different culture media (Miranda & Clement 1990).…”
