Stomatal analysis of citrus somatic hybrids obtained by protoplast fusion

Abstract: -The objective of this work was to evaluate leaf epidermis morphological characteristics of three citrus somatic hybrids, compared to their parents. Parental and somatic hybrid young leaves were collected and processed for scanning electron microscope observations. Citrus polyploid hybrids have fewer stomata per area and these are larger compared to their diploid parental parents. No differences in internal arrangement of the stomatal cells were detected between parental plants and somatic hybrids. Additional … Show more

“…Scanning electron microscope analysis has been previously used as an effective method to confirm protoplast fusant. Costa et al (2004) applied the SEM analysis to evaluate leaf epidermis morphological characteristics of three citrus somatic hybrids, compared to their parents. Although morphological feature can be signs for primary identification of the fusants, the DNA based genetic markers could provide the detailed information of the nuclear constitution and genetic relationship between the fusant and its parents.…”

“…Costa et al (2004) applied the technology to evaluate leaf epidermis morphological characteristics of three citrus somatic hybrids, compared to their parents. Random amplification polymorphic DNA (RAPD), which scans numerous loci in the genome is a useful genetic marker method (Welsh and McClelland 1990;Williams et al 1990).…”

“…Examinando estatisticamente os dados (Tabela 2), utilizando análise de variância (ANOVA) a três fatores (cultivar, tempo e concentração), conclui-se que existe diferença significativa entre as cultivares (p = 0,039 (GANTAIT et al, 2011). A técnica mais precisa na análise de estômatos utiliza microscópio eletrônico de varredura (COSTA et al, 2004). Entretanto, a disponibilidade do equipamento limita o seu uso em determinados grupos de pesquisa.…”
