ABSTRACT. Analysis of DNA polymorphisms allows for the genetic identification and precise discrimination of species with a narrow genetic base such as common bean. The primary objectives of the present study were to molecularly characterize commercial common bean varieties developed at various research institutions using microsatellite markers and to determine the degree of genetic diversity among the bean varieties analyzed. Fifty cultivars representing 12 grain classes and 64 genitors, i.e., accessions used to develop these cultivars, were characterized. Based on an analysis of 24 simple sequence repeats, the estimates for the average number of alleles and genetic diversity were 8.29 and 0.646, respectively. The combined probability of identity was estimated at 7.05 x 10 -17 , indicating a high individual discriminatory power. Thirtytwo percent of the cultivars exhibited heterogeneity for multiple loci that reflected either homozygosity for different alleles of a given locus in different individuals or heterozygosity for the locus. The average 1964-1978 (2014) Molecular characterization of cultivated common bean genetic diversity for the groups of cultivars and genitors was 0.605 and 0.660, respectively, with no genetic differentiation (F ST ) between these groups. Although similar estimates of expected heterozygosity were observed when the cultivars were grouped by release date, a greater number of private alleles was observed in the most recent cultivars. The genetic differentiation among cultivars originating from different institutions was not different from zero (F ST = 0.01). The molecular profile database derived from these analyses may increase the statistical power of genetic estimates and may be incorporated into breeding programs for common bean. Furthermore, the profiles obtained for the different cultivars may be used as molecular descriptors to complement traditional descriptors used in distinctiveness, uniformity and stability tests, thereby improving the traceability of samples and their derivatives and helping to protect the intellectual property rights of breeders.

-The objective of this work was to identify single-nucleotide polymorphism (SNP) markers linked with quantitative trait loci (QTLs) associated with increased contents of protein and oil in soybean. A total of 169 Brazilian soybean varieties, genotyped with 6,000 SNP markers, were evaluated. Protein and oil contents were obtained with the near-infrared reflectance method. Correlation and multiple linear regression analyses were used to identify linkage disequilibrium between SNP markers and the QTLs associated with the two characteristics. Seven QTLs were found to be associated with protein content, on six chromosomes (2, 6, 11, 12, 13, and 16), explaining 60.9% of the variation in this trait. For oil content, eight QTLs were identified on six chromosomes (1, 4, 5, 6, 17, and 19), explaining 78.3% of the variation in the trait. The correlation between the number of loci containing favorable alleles and the evaluated characteristics was 0.49 for protein content and 0.60 for oil content. The molecular markers identified are mapped in genomic regions containing QTLs previously mapped for both characteristics, which reinforces the association between these regions and the genetic control of oil and protein contents in soybean.Index terms: Glycine max, association genetics, genomic selection, grain quality, linkage disequilibrium, marker-assisted selection. Associação genômica ampla para mapeamento de QTLs ligados a conteúdos de proteína e óleo em sojaResumo -O objetivo deste trabalho foi identificar marcadores de polimorfismos de nucleotídeo único (SNPs) ligados a locos controladores de características quantitativas (QTLs) associados a maiores conteúdos de proteína e óleo em soja. Foram avaliadas 169 cultivares brasileiras de soja, genotipadas com 6 mil marcadores SNPs. Os conteúdos de proteína e óleo foram obtidos pelo método de refletância no infravermelho próximo. As análises de correlação e regressão linear múltipla foram utilizadas para a identificação de desequilíbrio de ligação entre os marcadores SNPs e os QTLs associados às duas características. Foram identificados sete QTLs associados ao conteúdo de proteína, em seis cromossomos (2, 6, 11, 12, 13 e 16), o que explicou 60,9% da variação nesta característica. Para o conteúdo de óleo, foram identificados oito QTLs em seis cromossomos (1, 4, 5, 6, 17 e 19), o que explicou 78,3% da variação na característica. A correlação entre o número de locos com alelos favoráveis e as características avaliadas foi de 0,49 para o conteúdo de proteína e de 0,60 para o conteúdo de óleo. 1RESUMO -Marcadores microssatélite têm sido utilizados na determinação da pureza genética em programas de melhoramento e de produção de sementes, visando garantir a qualidade genética das sementes. Objetivou-se neste trabalho demonstrar, com auxílio de marcadores moleculares microssatélites, que variações na coloração do hilo em sementes de soja, muitas vezes não constituem variação genética. Foram avaliadas sementes de soja da cultivar CD 222 que apresenta sementes com hilo preto, e de duas linhagens (CD 02RV-8444 e CD 01RV-7618), as quais apresentam sementes com hilo marrom. As sementes com variações na coloração e tonalidade do hilo encontradas em cada genótipo foram separadas visualmente em grupos. O DNA de cada semente foi extraído e analisado com marcadores microssatélites em "bulks" contendo DNA de cinco sementes. As variações encontradas na coloração do hilo da cultivar CD 222 e da linhagem CD 02RV-8444 não foram devidas à variação genética, considerando 16 locos microssatélites. Para a linhagem CD 01RV-7618, os bulks de sementes com hilo marrom pigmentado de preto (B1), com hilo marrom mais intenso (B2) e intermediário (B3), foram considerados iguais. Para esta mesma linhagem foi detectada diferença no bulk de sementes com hilo marrom menos intenso (B4), com o primer Satt 070. Duas sementes deste bulk apresentaram o mesmo padrão molecular observado para os outros bulks, no mesmo loco. As demais sementes foram consideradas misturas. A utilização de marcadores microssatélites demonstra que variações na cor ou tonalidade do hilo da soja nem sempre correspondem à variação genética.Termos para indexação: Glycine max, cor do hilo, pureza genética, SSR.
