Abstract: The interleukin (IL)-2R alpha chain (CD25) is expressed on regulatory T cells (Treg), which constitute more than 85% of the CD25+ T cell population in a naïve mouse. CD25 is also expressed on effector T cells in mice suffering from an acute infection by the obligate intracellular protozoan parasite, Toxoplasma gondii. Lethal toxoplasmosis is accompanied by a significant loss of Treg in mice naturally susceptible to toxoplasmosis. The present study was done to explore the role of Treg cells using an anti-CD25 a… Show more

“…For cytokine detection, splenocytes were recovered and purified as described previously (13,21) and stimulated for 72 h with 10 g/ml Toxoplasma extract (TE) or with purified anti-CD3 (clone 145-2C11) (eBioscience) at 1 g/ ml. The cells (5 ϫ 10 5 ) were seeded into 24-well plates in 1 ml RPMI 1640 containing 5% fetal calf serum (FCS), and supernatants were collected at 24, 48, and 72 h after activation.…”

“…Cells were washed once in PBS and counted to determine total viable cell numbers by trypan blue exclusion. Standard procedures were used to stain 2 ϫ 10 5 to 5 ϫ 10 5 PECs and 10 6 splenocytes in 5% FCS in PBS and mouse Fc block (BD Biosciences) as previously described (21). Antibodies for the detection of CD4 (GK1.5), CD8 (eBioH35-17.2), CD25 (PC61), CD69 (H1.2F3), Ki-67 (SolA15), and Foxp3 (FJK-16S) were purchased from eBioscience.…”

“… 3 , 4 In response to specific intracellular signals, various pro-inflammatory (IL1β, IL12, IL18, TNFα, IFNγ) and anti-inflammatory (IL4, IL10, TGFβ) cytokines ( Table 1 ) give rise to a cytokinome that acts for specific immunological stimulus to develop immune response for susceptibility or resistance ( Figure 1 ) to toxoplasmosis. 4 7 …”

“…3,4 In response to specific intracellular signals, various pro-inflammatory (IL1β, IL12, IL18, TNFα, IFNγ) and anti-inflammatory (IL4, IL10, TGFβ) cytokines (Table 1) give rise to a cytokinome that acts for specific immunological stimulus to develop immune response for susceptibility or resistance (Figure 1) to toxoplasmosis. [4][5][6][7] Immunological studies on human infections have clearly concluded that cell-mediated immunity and IFNγ are paramount in the control of any infection particularly caused by the intracellular pathogen (T. gondii). 8 The inability of the humoral response alone (antibodies) to prevent Toxoplasma reactivation is evident by the fact that most of the HIVinfected people lack effective immunity hence exhibit symptoms of T. gondii infection and reactivation, even in the presence of high titers of specific IgG.…”
