Abstract: Monoclonal antibodies (mAbs) and human sera from gametocyte carriers were applied in the bio-assay to test for their transmission-blocking capacity. Competition ELISA's have been developed for the detection of natural transmission blocking antibodies. Approximately 55% of the sera blocking in the bio-assay gave positive results in these competition ELISA's.

“…As in previous studies (6,9,18,23,26,43,44), we found evidence for naturally acquired immunity against P. falciparum sexual stages. Overall, 22 to 28% of our samples were positive for sexual stage-specific antibodies, while 54 to 65% were positive for preerythrocyte-and asexual blood stage-specific antibodies.…”

“…These antigens have a role in the fertilization or sporogonic development of malaria parasites in mosquitoes (43,44). A proportion of gametocytes die in the human host without being passed on to a mosquito, thereby exposing sexual stage antigens to the human immune system.…”

“…Pfs230 antibody prevalence as detected by IgG1 and competition ELISA is 25-60% and more prevalent than anti-Pfs48/45 antibodies (10-40%) in endemic sera [14,63,76,78]. Seroreactivity against both antigens is higher when measured by immunoprecipitation [48,73,77,79].…”

“…Unprotected by the red blood cell, parasites are exposed for several hours to blood products that have been co-ingested with the blood meal [11]. Specific antibodies, complement and granulocytes have been shown to decrease or abrogate the sporogonic development [12][13][14][15][16].…”

“…Indirect evidence comes from studies showing that mosquito infection rates tend to increase in the field-based DMFA when autologous serum is replaced by naive serum [25-27, 45, 46]. The use of SMFA has formally demonstrated that whole serum and (now more common) purified IgG from malariaexposed individuals can reduce mosquito infection rate and density [32,47,48]. The use of purified IgG has the advantage that the transmission modulating effect of antibodies of this class of immunoglobulins can be examined independent of other serum components such as antimalarial drugs [49].…”
