Random amplified polymorphic DNA (RAPD) analysis of Lutzomyia longipalpis laboratory populations

Abstract: The phlebotomine sand fly Lutzomyia longipalpis has been incriminated as a vector of American visceral leishmaniasis, caused by Leishmania chagasi. However, some evidence has been accumulated suggesting that it may exist in nature not as a single but as a species complex. Our goal was to compare four laboratory reference populations of L. longipalpis from distinct geographic regions at the molecular level by RAPD-PCR. We screened genomic DNA for polymorphic sites by PCR amplification with decamer single primer… Show more

“…Their first application to the systematics of Phlebotomine sandflies was done by pioneers in the 1990's (Adamson et al, 1991;Booth et al, 1991;Adamson et al, 1993;Maingon et al, 1993;Zeledon et al, 1993;Booth et al, 1996;Esseghir et al, 1997;Friedrich and Tautz, 1997;Depaquit et al, 1998;Dias et al, 1998). They constitute powerful tools to define populations with evident consequences to characterise vectors and non-vectors, to emphasise cryptic species, to associate males with females in a same species or to propose evolutionary systematics.…”

“…Other variations in populations of L. longipalpis from Central and South America have been reported, such as behavioral, morphological and biochemical characteristics (Dias et al 1998, Lampo et al 1999, Araki et al 2009, and the existence of reproductive isolation among populations from Central and South America mediated by different sex pheromones has been already suggested (Ward et al 1988).…”

“…111 This vector may exist as a species complex with different genetic profiles and biologic properties. [112][113][114][115][116][117] Lutzomyia longipalpis is typically a peri-domestic phlebotomine sand fly in dry regions. American visceral leishmaniasis has not been associated with forested regions.…”
