Reduced schedule of human anti-rabies immunization with Fuenzalida & Palacios vaccine: additional data

Abstract: It was reevaluated a reduced schedule for anti-rabies post-exposure immunization with newborn mice nervous tissue vaccine (Fuenzalida 8c Palacios) in a group of 30 non exposed volunteers. The vaccine was administered by intramuscular injections on days zero, 2, 4, 16 and 27, in the deltoid area. Antibody levels were determinated by a simplified serum neutralization microtest on days zero, 16 and 37. On days 16 and 37 the antibody levels of the whole group was >0.5 IU/ml and >1.0 IU/ml, respectively. The … Show more

“…Zanetti et al izing antibody (VNA) titers <0.5 IU (4). These results contrasted with others obtained in well-controlled trials using the same regimen (5,6). Several reasons, such as improper transportation, storage or administration of the vaccine (e.g., injections into the gluteus instead of the deltoid muscle), variable NIH potency among vaccine batches, and other local problems, were proposed as possible causes of unsatisfactory antibody titers.…”

“…At 48 h after parturition, blood from 30 prime-vaccinated and 30 revaccinated dams and 60 offspring was collected, and the serum neutralizing antibody (SNA) titers were analyzed by a rapid focus fluorescent inhibition test (RFFIT) using serial dilutions 1:10 to 1:640 of serum samples and positive and negative serum controls in microplates. The plates were stained with fluorescein isothiocyanate (FITC)-labeled antirabies immunoglobulin (rabies conjugate; Fujirebio) and the titer of a standard reference serum diluted was determined in each test (8,22). The conventionally defined SNA titer of 0.5 IU/ml for humans was considered a cutoff for rabies immunization (1).…”

“…Embora a resposta mediada por células seja reconhecidamente importante para a proteção, esta atua em conjunto com a resposta humoral; e a maioria das vacinas licenciadas, se baseia na indução da produção de anticorpos séricos para comprovar sua eficácia (Plotkin, 2001 (Favoretto et al, 1993;Nicholson, Turner e Aoki, 1978;Roumiantzeff et al, 1988;Strady et al, 2001;Zanetti et al, 1989).…”
