The aim of this study was to analyse the density of astrocytes and neurons in the motor areas of the cortex, striatum and hippocampus of rats that performed exercise before or after cerebral ischemia. A total of 36 Wistar rats were separated into two groups: One that was subjected to Middle Cerebral Artery Occlusion (MCAO) and the other that underwent surgical access in the absence of ischemia. Both groups were subdivided into: Animals that underwent exercise prior to MCAO and sham surgery (AI and AC, n = 12); animals that underwent exercise after MCAO and sham surgery (DI and DC, n = 12); and animals that did not undergo exercise (SI and SC, n = 12). The brains of mice were sectioned and stained by the Nissl method for staining of neurons and the astrocytes were labeled by immunohistochemistry with glial fibrillary acidic protein. Physical training was conducted after the surgery for 6 weeks (30 min/day), 5 days per week. The animals exercised before cerebral ischemia showed greater density of neurons in the striatum, in the apex of the dentate gyrus of the hippocampus, in the motor cortex contra lateral to the lesion and more astrocytes in the motor cortex of the cerebral hemispheres compared with the untrained group (p<0.05). The density of neurons in the brain regions analysed remained unchanged in exercised animals from the fourth day after cerebral ischemia when compared with the sedentary group.

RESUMOIntrodução: a encefalopatia hipóxico isquêmica neonatal é uma doença devastadora para o cérebro do recém-nascido. Nas últimas duas décadas, as pesquisas experimentais trouxeram grande avanço nos conhecimentos fisiopatológicos. Para a integridade neuronal, é necessária energia suficiente para a célula manter o equilíbrio iônico. A hipotermia tem sido estudada como um método com alto grau de neuroproteção na hipóxia-isquemia (HI). Objetivo: verificar a potencialidade terapêutica da hipotermia como tratamento da encefalopatia hipóxico-isquêmica em recém-nascidos. Método: foram consultadas as bases de dados do Medline, Lilacs, Scielo e JCR-ISI. As consultas incluíram artigos registrados entre 1993 e 2008 no idioma Português, Inglês e Espanhol. Discussão: encefalopatia hipóxico-isquêmica neonatal é complicação imediata à asfixia grave e pode causar graus variados de dano cerebral. As características do dano hipóxico isquêmico indicam que existe um período intermediário, em que é possível intervir interrompendo a cadeia de eventos que levam a destruição celular definitiva. Com a finalidade de proteger o cérebro dos insultos isquêmicos, utilizam-se drogas e diferentes procedimentos, tais como manitol, removedores de radicais livres, antagonistas de receptores opiáceos, supressores de metabolismo e hipotermia para minimizar as lesões. Conclusão: o resfriamento cerebral é a conduta terapêutica promissora em reduzir danos cerebrais em recém-nascidos. Objective: To evaluate the neuroprotection of mild hypothermia, applied in different moments, in temporary focal cerebral ischemia in rats. Methods: Rats was divided into Control (C), Sham (S), Ischemic-control(IC), Pre-ischemic Hypothermia (IH1), Intra-ischemic Hypothermia (IH2), and Post-ischemic Hypothermia (IH3) groups. Morphometry was performed using the KS400 software (Carl Zeiss IH3 and IH1 vs. IH2 (p=0.57; p=0.79). Conclusion: Pre-ischemic and intra-ischemic hypothermia were shown to be similarly neuroprotective, but this was not true for post-ischemic hypothermia.Key words: brain ischemia, mild hypothermia, morphometry, neuroprotection, reperfusion. RESUMOObjetivo: Avaliar a neuroproteção da hipotermia leve, aplicada em diferentes momentos, durante isquemia cerebral focal temporária em ratos .Métodos: Ratos foram divididos em grupos: Controle (C), Sham (S), Controle-isquêmico (IC), Hipotermia Pré-isquêmica (IH1), Hipotermia Intra-isquêmica (IH2) e Hipotermia Pós-isquêmica (IH3). A morfometria foi realizada em secções coronais coradas por Luxol Fast Blue através do programa KS400 (Carl Zeiss ® ). Foram calculados áreas e volumes isquêmicos. Resultados: Estatisticamente, áreas azuis demonstraram diferença entre os grupos C vs. IC, IC vs. IH1 e IC vs. IH2 (p=0,0001; p=0,01; p=0,03), e nenhuma diferença entre C vs. S, IC vs. IH3 e IH vs. IH2 (p=0,39; p=0,85; p=0,63). Áreas vermelhas demonstraram diferença entre C vs. IC, IC vs. IH1 e IC vs. IH2 (p=0,0001; p=0,009; p=0,03), e nenhuma diferença entre C vs. S, IC vs. IH3 e IH1 vs. IH2 (p=0,48; p=0,27; p=0,68). Áreas isquêmicas médias e volumes isquêmicos demonstraram diferença entre os grupos IC vs. IH1 e IC vs. IH2 (p=0,0001 and p=0,0011), e nenhuma diferença entre IC vs. IH3 and IH1 vs. IH2 (p=0,57; p=0,79). Conclusão: Hipotermias pré-isquêmica e intra-isquêmica demonstraram neuroproteção em grau semelhante, o que não ocorreu com hipotermia pós-isquêmica.Palavras-Chave: isquemia encefálica, hipotermia leve, morfometria, neuroproteção, reperfusão.
