Protein biomarkers of external root resorption: A new protein extraction protocol. Are we going in the right direction?

Abstract: OBJECTIVE: The aim of this study is to determine a protocol of gingival crevicular fluid protein extraction used for the first dimension of 2-DE gels. It also aims at conducting a review on the current candidates for protein markers of this pathology, all of which may be used to prevent the disease. METHODS: Gingival crevicular fluid was collected from two groups of 60 patients each, with and without external root resorption. Samples were extracted by means of various methods of protein extraction. SDS-PAGE ge… Show more

“…19 A total of 9 studies examined the resorption severity grades measured in mm, or classified it as mild/moderate/severe, or as coronal/apical resorption. 5,7,9,12,13,19,20,24,29 Resorption with respect to the duration of orthodontic treatment was considered in 7 studies. 7,9,19,20,23,27,30 While the majority of studies had both male and female participants, two studies investigated only female participants.…”

“…GCF biomarker detection has already been employed to clinically track osteoclast activity, bone remodeling, and ERR occurring in particular during orthodontic treatment [ 31 ]. George and Evans tested the hypothesis that during root resorption organic proteins and cytokines from the surrounding bone and dentin are released into the gingival crevice.…”

“…To achieve this goal, there is still a need for more highresolution protein research methods to look for new molecular markers. The technique of choice for this task is one-dimensional SDS-PAGE, as a proteinextract quality evaluation step followed by twodimensional electrophoresis (2DE) and mass spectrometry 10 . Thus, this study attempts to optimize the method of GCF collection from healthy gingival sites, storage and total protein extraction method that is suitable for the highthroughput 2DE gel-based proteomic analytical techniques.…”
