Background: Cephaelis ipecacuanha is a medicinal plant used in folk medicine for the treatment of amebiasis. However, the pharmacological properties of this species remain poorly understood. Objective: Characterize chemical composition and in vitro antiprotozoal activity of Cephaelis ipecacuanha. Method: The aqueous extract was lyophilized using the Chisrt Alpha apparatus and transferred to 96-well plates at concentrations ranging from 62.5 to 1000 μg / mL. The antiprotozoal activities against Trypanosoma cruzi, Leishmania brasiliensis, and L. infantum were evaluated after 72h incubation. Fibroblasts incubated at the same conditions for 24h were used as a cytotoxicity control. The readings were performed by spectrophotometry after staining with resazurin. The chemical composition of the extract was analyzed by High-Performance Liquid Chromatography (HPLC-DAD). Results: The results demonstrated that C. ipecacuanha had moderate antiprotozoal activity against T. cruzi. However, its cytotoxicity against the fibroblasts was significantly higher. Conclusion: This finding suggests that the use of this plant by the population, besides not having significant benefits, can cause associated health risks.

Duroia longiflora (Rubiaceae) is an Amazonian plant species with no previous chemical study. The aim of this work was to study the chemical composition of D. longiflora leaves and study their histochemistry. In vivo tissues of the leaves were histolocalized. Extracts were made from dried leaves with hexane, methanol, and water and the methanolic extract was fractionated. Fractionation of the dichloromethane phase of the methanolic leaf extract allowed the isolation of a biflavanoid. Via nuclear magnetic resonance (mono and bidimensional), mass spectrometer analysis, and comparison with existing literature, its chemical structure was identified as 2α,3α-epoxy-5,7,3′,4′-tetrahydroxyflavan-(4β→8)-epicatechin. This is the first report describing the histochemical and phytochemical studies of D. longiflora leaves. As far as we know, this is the first report of this substance in any Rubiaceae species.

Duroia saccifera (Rubiaceae) occurs in the Amazon rainforest and their extracts showed antibacterial properties. To obtain greater quantities of active substances, leaf segments from in vitro D. saccifera seedlings were used as explants for calli induction; calli were multiplied via multiple subcultures, dried and extracted with hexane followed by ethyl acetate (EtOAc) and methanol (MeOH). As D. macrophylla had been reported to produce antimycobacterial substances, we assayed calli extracts against Mycobacterium tuberculosis (H37Rv strain). Calli EtOAc extract was active, with a minimal inhibitory concentration (MIC) of ≤ 25 mg mL-1, IC90of 19.5 mg mL-1 and minimal bactericidal concentration (MBC) of 200 mg mL-1. EtOAc extract was analyzed by Thin Layer Chromatography (TLC) and Nuclear Magnetic Resonance (NMR) to determine its chemical profile, and was found to be rich in terpenes. Chromatographic fractionation of the EtOAc extract yielded a mixture of two sterols, β-sitosterol and stigmasterol (in proportion of 2:1), which were identified by 1H and 13C NMR analysis. As far as we know, this is the first report of Duroia saccifera in vitro cell culture, antituberculosis activity of calli extract and β-sitosterol and stigmasterol isolation from in vitro plant cell culture.
