Animal models of alcohol and drug dependence

Abstract: Drug addiction has serious health and social consequences. In the last 50 years, a wide range of techniques have been developed to model specific aspects of drug-taking behaviors and have greatly contributed to the understanding of the neurobiological basis of drug abuse and addiction. In the last two decades, new models have been proposed in an attempt to capture the more genuine aspects of addiction-like behaviors in laboratory animals. The goal of the present review is to provide an overview of the preclini… Show more

“…The modus operandi for drug use in 41 animals is the self-administration paradigm (18,865 publications between 2009-42 2019 via PubMed), in which, animals perform a behavior to consume a drug or 43 saline. Numerous variations of drug self-administration exist to model addiction 44 related behavior, but our study predominately focuses on the classic (short-access) 45 cocaine self-administration paradigm, which is considered a valid model for 46 reinforcement learning and voluntary cocaine use 2,3 . These features seem important 47 for the development of substance abuse, but controversy remains pertaining to the 48 relationship between classic animal self-administration paradigms and human drug 49 addiction 4,5 .…”
