The present study determined the expression and biological functions of FOXA2 gene in colon cancer in tissues, cells and animals. A total of 66 patients with colon cancer were included in the present study. Using The Human Protein Atlas database, expression and distribution of FOXA2 in colon cancer tissues were analyzed. Using immunohistochemistry, the expression and distribution of FOXA2 in colon cancer cells were studied. Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) was performed to determine the expression of FOXA2 mRNA in colon cancer tissues. Following in vitro transfection with FOXA2 interference sequence (siR-FOXA2), the proliferation, cell cycle, migration and invasion of colon cancer HCT116 and HT29 cells were investigated using Cell Counting Kit-8 assay, flow cytometry, and Transwell assay, respectively. Furthermore, flow cytometry was used to determine apoptosis of HCT116 and HT29 cells. Western blotting was used to determine the expression of epithelial mesenchymal transition (EMT) proteins, E-Cadherin and Vimentin. Laser scanning confocal microscopy was performed to observe the cytoskeleton in HCT116 and HT29 cells. Results indicated tumorigenesis of colon cancer cells in nude mice. In addition, the expression of FOXA2 in colon cancer tissues was elevated and associated with the metastasis and clinical staging of colon cancer. Notably, inhibition of FOXA2 reduced the proliferation of colon cancer cells in vitro and reduced expression of FOXA2 was able to decrease the migration and invasion abilities of colon cancer cells. Furthermore, FOXA2 promoted EMT, inhibited apoptosis and enhanced the invasion ability of colon cancer cells. Decreased expression of FOXA2 inhibited tumorigenesis of colon cancer cells in nude mice. To conclude, the present study demonstrated that the expression of FOXA2 in colon cancer tissues was elevated and associated with the metastasis and clinical staging of colon cancer. As an oncogene, FOXA2 may promote the proliferation, migration and invasion and EMT in colon cancer.

Objective The study aimed to characterize the clinical, histological, and immunohistochemical profile of women with invasive breast cancer, according to the risk for Hereditary Predisposition Breast and Ovarian Cancer Syndrome in a Brazilian population. Methods This is a retrospective study performed from a hospital-based cohort of 522 women, diagnosed with breast cancer treated at an oncology referral center in the Southeast region of Brazil, between 2014 and 2016. Results Among the 430 women diagnosed with invasive breast cancer who composed the study population, 127 (29.5%) were classified as at increased risk for hereditary predisposition to breast and ovarian cancer syndrome. There was a lower level of education in patients at increased risk (34.6%) when compared with those at usual risk (46.0%). Regarding tumor characteristics, women at increased risk had higher percentages of the disease diagnosed at an advanced stage (32.3%), and with tumors > 2cm (63.0%), with increased prevalence for both characteristics, when compared with those at usual risk. Furthermore, we found higher percentages of HG3 (43.3%) and Ki-67 ≥ 25% (64.6%) in women at increased risk, with prevalence being about twice as high in this group. The presence of triple-negative tumors was observed as 25.2% in women at increased risk and 6.0% in women at usual risk, with the prevalence of absence of biomarkers being 2.5 times higher among women in the increased risk group. Conclusion From the clinical criteria routinely used in the diagnosis of breast cancer, the care practice of genetic counseling for patients at increased risk of hereditary breast cancer in contexts such as Brazil is still scarce.

O câncer de mama é a neoplasia maligna mais comum entre as mulheres no Brasil, sendo uma das principais causas de morte por câncer feminino. A avaliação dos parâmetros prognósticos, como subtipo histológico e presença de biomarcadores, é fundamental, tendo a imuno-histoquímica um importante papel, com consequentes melhorias no tratamento. O presente estudo objetivou determinar o perfil histopatológico e molecular do câncer de mama em mulheres assistidas em centro de oncologia. Trata-se de um estudo documental, analítico e retrospectivo, que utilizou prontuários de mulheres com câncer de mamaatendidas em um centro de oncologia do Agreste Pernambucano, nos anos de 2014 a 2016. O estudo contou com 180 prontuários de pacientes com diagnóstico de câncer de mama. Observou-se uma prevalência de mulheres pardas (81,66%), casadas (53,33%) e com idade maior que 45 anos (81,66%). Com relação à classificação histopatológica, houve uma maior predominância do carcinoma ductal invasivo grau 2 (54,05%). Quanto ao subtipo molecular, o luminal B-HER2 negativo foi o mais frequente, representando 34,48% dos casos, seguido do luminal A, com 25%. O diagnóstico de carcinoma ductal invasivo foi mais prevalente na idade entre 51 e 60 anos (26,3%) e grande parte das mulheres com este diagnóstico (35,05%)possuíam o subtipo molecular luminal B-HER2(-). Os dados obtidos revelaram que a idade elevada, raça/cor parda, baixo nível de escolaridade, estado civil casada, exame histopatológico evidenciando carcinoma ductal invasivo e exame imuno-histoquímico revelando subtipo molecular luminal B-HER2(-) estavam entre as características mais frequentes nas mulheres com câncer de mama na região.
