Molecular detection of mixed infections with multiple dengue virus serotypes in suspected dengue samples in Tamaulipas, Mexico

Abstract: This study aimed to detect dengue virus (DENV) serotypes in serum samples obtained in Matamoros Tamaulipas, Mexico, and to determine the concordance of conventional nested reverse transcriptase polymerase chain reaction (RT-PCR) and a serological test [enzyme-linked immunosorbent assay (ELISA NS1)]. Here, we detected mixed infections consisting of four serotypes of DENV. The most prevalent serotype was DENV-1, followed by DENV-4. This is the first report of DENV-4 in our region. Mixed infections were also dete… Show more

“…The DENV panel confirmed the serotypes of 12 RT-PCR-serotyped patient samples or clinical isolates (Fig 3F, Fig. S25), and identified 2 clinical isolates with mixed infection, a commonly observed phenomenon(18). SHERLOCK can therefore be extended to differentiate between related viruses or serotypes using a single amplification reaction.…”

“…Two earlier studies from India, including ours, and one from Mexico used this PCR for identifying viremia among serologically diagnosed patients with dengue. DENV RNA positivity was found to be 48.6% (53/109)and 52% (71/136) among NS1 ELISA positives or 59.5% among 117 NS1 and 4 IgM positives (immunochromatographic rapid test) . Overall, detection of DENV RNA among NS1‐positive patients with dengue was approximately 50% when CprM region–based PCR was used for screening.…”

These observations concur with the results reported earlier [94].…”
