Nullity of GSTT1/GSTM1 related to pesticides is associated with Parkinson's disease

Abstract: Parkinson's disease (PD) is a progressive neurodegenerative disorder, clinically characterized by bradykinesia, rigidity, resting tremor, and postural instability. Early symptoms of PD are primarily due to the selective degeneration of dopaminergic neurons of the substantia nigra, innervating the neostriatum. The primary cause of PD is unknown, however, mitochondrial failure, oxidative stress and genetic factors, responsible for neurodegeneration in the substantia nigra are investigated hypotheses for the etio… Show more

“…This is an agreement with previous report of Kubiszeski et al [9] in Cuiabá population (50% and 32%) and Rocha et al [10] in Amazônia population. Other studies in the Brazilian population from Bahia, performed by Rocha et al [10] and Pinhel et al [11] in the population from São Paulo confirm our findings in the Goiânia population. GSTM1 and GSTT1 enzymes metabolize several precarcinogens, drugs, constituents of tobacco smoke and solvents to reactive metabolites which ultimately lead to DNA or protein damage [12].…”

“…Alterations in the expression of GST isoenzymes in brain have been connected to neurological disorders including Alzheimeŕs disease [142], epilepsy [143] and Parkinsońs disease [144,145]. However, it remains to be elucidated whether the expression or activity of astrocytic GSTs are altered in such diseases.…”

“…Furthermore, other genes re1ated to PD, such as APOE, GSTs and LRRK2 (PARK8), also showed no difference between patients and controls in this population 20,21,22,23 . On the other hand, mutations in the genes GIGYF (PARK11), ATP13A2 (PARK9) and GBA were associated with PD 6 .…”
