Abstract: Objective: We sought to test the effect of different dosages of pioglitazone (PIO) on the glomerular expression of podocalyxin and urinary sediment podocalyxin excretion and to explore the potential renoprotective mechanism. Materials and methods: Type 1 diabetes induced with streptozotocin (65 mg/kg) in 36 male Sprague-Dawley rats were randomly allocated to be treated with vehicle or 10, 20, 30 mg/kg/d PIO respectively for 8 weeks. Eight rats were enrolled in the normal control group. Results: At 8 th week, r… Show more

“…PODXL1 is an anti-adhesive transmembrane protein that has been reported to be expressed on the foot processes of podocytes in the kidney glomerulus, as well as on the endothelium at certain sites ( 36 ). The results of the present study revealed that PODXL1 expression was decreased in diabetic rats following STZ injection, which is consistent with the observations of a previous study ( 38 ).…”

“…Consistent with these observations, a previous study showed that pioglitazone treatment for 4 weeks had no effect on the serum HDL levels in Wistar-Kyoto rats 43 . On the other hand, Xing et al demonstrated that pioglitazone treatment for 8 weeks increased the plasma HDL levels in Sprague-Dawley rats 44 . Moreover, in a randomized clinical trial, pioglitazone treatment for 24 weeks was found to be associated with a decrease of the carotid intima-media thickness in type 2 diabetics, along with increase of the plasma HDL levels 45 .…”

“…PIO administration decreased urinary sediment podocalyxin excretion, preserved glomerular podocalyxin expression and restored normal function of most podocyte structure-related genes. It also suppressed apoptosis, hypertrophy and cell death, and preserved their differentiation [ 97 101 ].…”
