Detection of Yersinia pestis fraction 1 antigen with a fiber optic biosensor

Abstract: A fiber optic biosensor was used to detect the fraction 1 (F1) antigen from Yersinia pestis, the etiologic agent of plague. The instrument employs an argon ion laser (514 nm) to launch light into a long-clad fiber and measures the fluorescence produced by an immunofluorescent complex formed in the evanescent wave region. This sensing area is a short section (12.5 cm) at the end of the optical fiber from which the cladding has been removed and in which the silica core has been tapered. Capture antibodies, which… Show more

“…However, this method depends on subjective interpretation (visualization of the spots). Recently, Cao et al (1995) have described the detection of Y. pestis F1 antigen by using a fiber optic biosensor but it is still a sophisticated procedure to be employed in developing countries. Therefore, ELISA remains the most reliable method.…”

“…На основе оптоволокна были разработаны иммуносенсоры для выявления капсульного антигена «фракция 1» чумного микроба [10,11,14] и антител к нему [12], для обнаружения протективного антигена сибиреязвенного микроба [13]. Чувствительность биосенсоров составила 5-50 нг/мл белка [10], время отклика -1 мин [14]. Оптоволоконные биосенсоры нашли свое отражение в системе обнаружения ПБА RAPTOR™ (от англ.…”

“…The genes of pXO2 encode proteins involved in biosynthesis of the poly-D-glutamic acid capsule. Plague, caused by Y. pestis, is a zoonosis mainly circulating between rodents and their fleas, which can transmit the disease to humans (Cao et al 1995). The most important clinical forms of plague are bubonic, septicaemic and pneumonic.…”
