Abstract: Blood basophils are a target for VIT, which induces impaired release of both preformed and newly generated mediators. However, we believe the basic mechanisms of rapid clinical tolerance induced by ultra-rush VIT remain to be investigated.

“…Our data show that the effect of DNA immunization is dominant, since it prevents the subsequent induction by PLA 2 /alum of either an IgE Ab response or activation of Th2 cells producing IL-4; it can also reduce a pre-existing allergen-specific IgE response. “…Significant decreases of basophil histamine release were demonstrated after the maintenance dose was administered for several months, and a decrease of total histamine in peripheral blood leukocytes was observed just after completion of ultrarush or rush VIT, possibly reflecting a certain depletion of circulating basophils (82,83,85). However, in vitro histamine release from blood basophils in response to allergen stimulation was not significantly affected by rush or ultrarush VIT if the results were expressed as percent release of total histamine (85).
