Abstract: Brazzein is the smallest sweet-tasting protein and was isolated from the wild African plant Pentadiplandra brazzeana. The brazzein molecule consists of 54 amino-acid residues and four disulfide bonds. Here, the first crystal structure of brazzein is reported at 1.8 Å resolution and is compared with previously reported solution structures. Despite the overall structural similarity, there are several remarkable differences between the crystal and solution structures both in their backbone folds and side-chain co… Show more

Due to this structural conservation and their sequence similarity, defensins from arthropods and mollusks are predicted as evolutionarily related [28]; nevertheless, they [59], (i) AhPDF1 -PDB: 2M8B from Arabidopsis halleri [64], (j) PhD1 -PDB: 1N4N from Petunia hybrida [40], (k) dimeric conformation of NaD1 -PDB: 4AAZ from Nicotiana alata [55] and (l) the defensin-like Brazzein -PDB: 4HE7 from Pentadiplandra brazzeana [68], (m) sequence alignment of some classic CS␣␤ defensins from plants. “…Brazzein also contains four intramolecular disulfide bonds (Cys4‐Cys52, Cys16‐Cys37, Cys22‐Cys47 and Cys26‐Cys49), resulting in a structure with good stability at high temperature and wide pH ranges (Nagata et al . .…”

“…The following residues are absent in brazzein as part of its composition Methionine (Met), Threonine (Thr) and Tryptophan (Trp). The crystal structure of brazzein 4HE7 with 54 amino-acid residues as reported by [2] is given in Figure 8. The structure shows that the protein has one alphahelix and three strands of anti-parallel beta sheet linked by backbone hydrogen bonds and four equally spaced disulfide bonds that sustain the structure in a stable conformation.…”

“…Brazzein was isolated from the fruit of wild African plant Pentadiplandra brazzeana Baillon [1,2]. Brazzein has inherent sweetness 500 to 2000 times that of sucrose on a molar basis with reported different uses such as tastemodifying protein and as an alternative to low calorie sweeteners [3].…”
