Abstract: The epithelial lining of the small intestine consists of multiple cell types, including Paneth cells and goblet cells, that work in cohort to maintain gut health. 3D in vitro cultures of human primary epithelial cells, called organoids, have become a key model to study the functions of Paneth cells and goblet cells in normal and diseased conditions. Advances in these models include the ability to skew differentiation to particular lineages, providing a useful tool to study cell type specific function/dysfuncti… Show more

“…Simultaneous addition of DAPT and the Wnt pathway inhibitor IWP2 (ENR+DI), on the other hand, results in intestinal organoid cultures in which goblet cell differentiation is favored ( Figure 1B ; Yin et al., 2014 ). This system has been used to identify cell type-specific regulators of Paneth and goblet cells through transcriptomic analysis and comparison of ENR organoids to Paneth and goblet cell-enriched organoids ( Treveil et al., 2019 ). In a separate study, using single-cell RNA-sequencing (scRNA-seq) of ENR organoids, Paneth cell-enriched organoids, and freshly isolated primary mouse intestine tissue, researchers identified Nupr1 as a potentially important transcription factor for the survival and development of Paneth cells ( Mead et al., 2018 ).…”

“…Organoids allow directed differentiation towards both cell identities, yielding organoids heavily enriched in either goblet cells or Paneth cells [62]. Comparing transcriptomes of these skewed organoids helped identify key regulators of the differentiation pathway [63]. Further analysis of the omics data as well as functional analysis of these organoids will allow better understanding of the role of both cell types in innate immune defence.…”

“…Again, starting with human embryonic stem cell derived intestinal organoids possessing gene expression patterns that generated human duodenum or ileum depicting regional specification of the developing human intestine [67], these authors showed that active FGF and WNT signaling was associated with organoids which possessed gene expression patterns that generated human duodenum or ileum suggesting regional specification of the developing human intestine [67]. By analyzing regulatory networks of murine small intestinal organoids, [12] suggested that multifactorial mechanisms may be involved in genetic rearrangement which gives rise to inflammatory bowel disease.…”

“…These questions have occupied the minds of stem cell scientists for a long time. Brain [2][3][4], heart [5,6], intestine [7,8], skeletal muscle [9], stem cells have been identified and shown to play the functional role of tissue replacement and sustenance of specific tissue integrity [10][11][12] thereby providing tools for drug discovery and for studies on regenerative medicine [13]. For example, hope has been raised in the potential of stem cells [14] for use in therapy see reviews by [15][16][17][18].…”
