Abstract: In the last 20 years, severe wheat stem rust outbreaks have been recorded in Africa, Europe and Central Asia. This previously well controlled disease, caused by the fungus Puccinia graminis f. sp. tritici (Pgt), has re-emerged as a major threat to wheat cultivation. The stem rust (Sr) resistance gene Sr22 encodes a nucleotide-binding and leucine-rich repeat (NLR) receptor which confers resistance to the highly virulent African stem rust isolate Ug99. Here, we show that the Sr22 gene is conserved among grasses … Show more

“…A few of the identified R genes in wheat include Sr5 , Sr13 , Sr23 , Sr27 , Sr36 , Sr40 , etc., which have varying effect on Pgt races. In wheat, R gene Sr5 is known to limit the growth of avirulent Puccinia strain, while Sr22 advances the development of Pgt races ( Hatta et al, 2020 ; Wu et al, 2020 ). Various researchers mapped Sr genes on wheat chromosomes 1BS, 2B, 3B, 5DL, 6AL, 6DS, and 7AL ( Table 4 ), conferring resistance against stem rust during adult-plant stage.…”

“…TraesCS2A02G001500 was predicted to encode a protein with nucleotide binding sites and multiple leucine-rich repeats (NBS-LRR), the typical structures of disease resistance genes (R genes). Many cloned wheat rusts resistance genes are found to encode NBS-LRR proteins, including leaf rust resistance genes ( Lr1 , Lr10 , Lr21 , and Lr22 ) ( Feuillet et al, 2003 ; Huang et al, 2003 ; Hiebert et al, 2007 ; Qiu et al, 2007 ), stripe rust resistance genes ( Yr5 and Yr10 ) ( McGrann et al, 2014 ; Yuan et al, 2018 ), and stem rust resistance genes ( Sr22 , Sr33 , Sr35 , Sr45 , and Sr50 ) ( Saintenac et al, 2013 ; Periyannan et al, 2014 ; Casey et al, 2016 ; Saur et al, 2019 ; Md Hatta et al, 2020 ). Our results suggest that TraesCS2A02G001500 might be the candidate gene of Lr65 .…”

“…Sr22 was cloned and its product was shown to contain typical CC, NB, and LRR regions although the gene was shown to belong to a small gene family with three homologs [ 143 ]. Fourteen orthologs of the Sr22 gene were later reported both in T. boeoticum and other related species including T. aestivum although only one of them seemed to confer resistance to P. graminis [ 144 ].…”
