Abstract: Baculovirus inhibitors of apoptosis (IAPs) act in insect cells to prevent cell death. Here we describe three mammalian homologs of IAP, MIHA, MIHB, and MIHC, and a Drosophila IAP homolog, DIHA. Each protein bears three baculovirus IAP repeats and an N-terminal ring finger motif. Apoptosis mediated by interleukin 13 converting enzyme (ICE), which can be inhibited by Orgyia pseudotsugata nuclear polyhedrosis virus IAP (OpIAP) and cowpox virus crmA, was also inhibited by MIHA and MIHB. As MIHB and MIHC were able … Show more

“…[1][2][3][4][5] A hallmark of IAPs is the presence of one to three baculovirus IAP repeat (BIR) domains, through which the IAPs bind and thereby inhibit caspases. [6][7][8] Certain IAPs (such as mammalian proteins XIAP, CIAP1, CIAP2 and ML-IAP) also possess a C-terminal RING finger, which can act as E3 ubiquitinligases and might enable IAPs to ubiquitylate proteins that interact with them directly and target them for degradation by the proteasome.…”

“…Members in the IAP protein family are generally characterized by one or more copies of the so-called baculovirus IAP repeat and by a ring finger domain at their carboxyl terminus. 15 The members of IAP family are thought to inhibit the terminal effector caspases directly through baculovirus IAP repeat (BIR)-dependent recognition. 16 At variance with the other IAP protein members, survivin has recently been characterized by a single baculovirus IAP repeat and no ring finger.…”

“…Instead, EC1 contains several genes whose products attenuate activation of NFkB. These include IkB-a and -e, which both directly block NFkB entry to the nucleus; TNFAIP3, BIRC3 and IER3, which encode distinct proteins annotated as attenuating various steps in the TNF signaling pathway downstream of TRAF [22][23][24][25] . We confirmed IL18 induction of many transcript changes by quantitative PCR using KG1 RNA samples and RNA samples prepared from whole human blood treated with recombinant IL18 (Table 1).…”
