Abstract: A new method for quantitative phospholipase activity assays using mass spectrometry (MS) and a supported thin film consisting of a graphene oxide (GO)/carbon nanotube (CNT) double layer as a substrate for laser desorption ionization (LDI) has been developed. Phospholipids were very efficiently analyzed by LDI-time-of-flight (TOF) MS on the GO/CNT films, presumably because of the affinity of phospholipids for the GO/CNT surface. Therefore, the rate of lipid hydrolysis was conveniently measured using LDI-TOF mas… Show more

“…Particularly, preferential binding of single stranded nucleic acid (NA) on GO compared to double stranded NA was the most important principle in the fluorescent NA/GO utilized biosensors [20,21]. The principle has been used to develop NA sensors [12,13], small molecule sensors with aptamer [22,23] and DNA or RNA related enzyme assay platforms [24][25][26]. More recently, researchers started to extend the GO based systems to analyze biomolecules present in live cells by utilizing nano-sized GO (NGO) [27][28][29].…”

“…Moreover, GO-based monohybrid films were synthesized to increase the efficiency of LDI by immobilizing GO on a solid surface and then multiwalled carbon nanotubes(MWCNT) and gold nanostructure were incorporated subsequently [150,151]. The GO-MWCNT composite showed high potential for analysis of phospholipase activity by LDI-MS [152] along with other small molecules and tissue imaging [153]. The efficiency of LDI-MS using GO-MWCNT platform can be increased further by increasing the thickness and roughness of the surface [154].…”
