Abstract: While the ApoE ε4 allele is a known risk factor for mild cognitive impairment (MCI) and Alzheimer's disease, brain region specific effects remain elusive. In this study, we investigate whether the ApoE ε4 allele exhibits brain region specific effects in longitudinal glucose uptake among patients with MCI from the Alzheimer's Disease Neuroimaging Initiative (ADNI). Preprocessed FDG PET images, MRIs, and demographic information were downloaded from the ADNI database. An iterative reblurred Van Cittertiteration m… Show more

“…1 and Table 4, the statistical significance levels were improved by PET with PVC in testing the ROI FDG SUVR differences between AD and MCI groups in left and right hippocampus, CA2/3/ DG, and right CA1. This is consistent with our previous Alzheimer's Disease Neuroimaging Initiative studies in small ROIs including amygdala and entorhinal cortex, where the PET spatial resolution of 8-mm FWHM [28,29]. In the study, we also performed RBV (region-based voxelwise)-RVC (reblurred Van Cittert iteration) PVC algorithm [46].…”

“…18 F-FDG PET image spatial normalization and ROI 18 F-FDG SUVR calculation were described in our previous studies [28][29][30]. Briefly, all PET images were co-registered to matched T1-weighted 3D BRAVO MRI images.…”

“…Glucose hypometabolism, to a lesser extent in terms of magnitude or spatial distribution, was also observed in patients with mild cognitive impairment (MCI), a prodromal stage of AD, suggesting an early role in the course of disease [12]. An 84-months longitudinal study clearly demonstrated an ApoE4associated brain-region specific longitudinally declined glucose metabolism pattern in the context of MCI [14]. Such an early role is further supported by the finding of abnormally low rates of glucose metabolism in the vulnerable brain regions in young adults carrying apoE4 allele in their 20s, several decades before the possible onset of dementia [15].…”

“…It has been conjectured that the higher prevalence in females is a result of longer life span 8,25 . Alternatively, studies have alluded to sex-specific hormonal and metabolic changes that interplay with the onset and progression of AD dementia 6,16,26 . Sex also interacts with apoE isoform status, where females with the apoE4 isoform are at increased risk compared to males 27-29 .…”
