Streptocaulon juventas is a well-known plant that has antimicrobial activity, in vitro antiplasmodial activity, anti-proliferative activity, and antioxidant activity. In this study, we showed experimental evidence that proved that S. juventas root ethanolic extract has wound healing activities. First, in a mouse excision wound model, S. juventas root ethanolic extract at a dose of 100 mg/kg/day significantly reduced the wound closure time. After 7 days, the wound granulation tissue in mice treated with the extract exhibited a 2.3-fold decrease in inflammatory cells, a 1.7-fold increase in fibroblasts and enhanced angiogenesis. Molecular analysis also revealed that after wounds were treated with S. juventas root ethanolic extract, TNF-α and NF-κB1 gene expression were down-regulated by 4.7 and 3.7 times, respectively. In contrast, TGF-β1 and VEGF gene expression were up-regulated by 1.9 and 6.5 times, respectively. Taken together, our experimental data strongly show that the ethanolic extract from S. juventas root displays remarkable wound healing activity.

Antibacterial and Antiviral Activities of Local Thai Green Macroalgae Crude Extracts in Pacific White Shrimp (Litopenaeus vannamei)

Macroalgae are potentially excellent sources of bioactive secondary metabolites useful for the development of new functional ingredients. This study was conducted to determine the antimicrobial efficacy of the hot water crude extracts (HWCEs) of three species of local Thai green macroalgae Ulva intestinalis (Ui), U. rigida (Ur), and Caulopa lentillifera (Cl) and a commercial ulvan from U. armoricana (Ua). Chemical analysis indicated that the HWCE of Ur showed the highest sulfate content (13.9% ± 0.4%), while that of Ua contained the highest uronic acid and carbohydrate contents (41.47% ± 4.98% and 64.03% ± 2.75%, respectively), which were higher than those of Ur (32.75% ± 1.53% and 51.02% ± 3.72%). Structural analysis of these extracts by Fourier-transform infrared (FTIR) spectroscopy revealed that these HWCEs are complex with a signal at 1250 cm−1 corresponding to S=O stretching vibrations, while the signals at 850 cm−1 were attributed to the C–O–S bending vibration of the sulfate ester in the axial position. These HWCEs showed the growth suppression against some pathogenic Vibrio spp. Interestingly, the HWCEs from Ui at concentrations of 5 and 10 mg/mL completely inhibited white spot syndrome virus (WSSV) in shrimp injected with HWCE–WSSV preincubated solutions. This inhibitory effect was further confirmed by the reduction in viral loads and histopathology of surviving and moribund shrimp.

Objective:This study evaluated in vitro cell viability by the colorimetric MTT stands for 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (3-(4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide) (3-(4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide) assay compared to image analysis by CellProfiler® software.Materials and Methods:Hepatoma (Hepa-1c1c7) and fibroblast (L929) cells were exposed to isolated substances, camptothecin, lycorine, tazettine, albomaculine, 3-epimacronine, trispheridine, galanthine and Padina gymnospora, Sargassum sp. methanolic extract, and Habranthus itaobinus Ravenna ethyl acetate in different concentrations. After MTT assay, cells were stained with Panotic dye kit. Cell images were obtained with an inverted microscope equipped with a digital camera. The images were analyzed by CellProfiler®.Results:No cytotoxicity at the highest concentration analyzed for 3-epimacronine, albomaculine, galanthine, trispheridine, P. gymnospora extract and Sargassum sp. extract where detected. Tazettine offered cytotoxicity only against the Hepa1c1c7 cell line. Lycorine, camptothecin, and H. itaobinus extract exhibited cytotoxic effects in both cell lines. The viability methods tested were correlated demonstrated by Bland–Atman test with normal distribution with mean difference between the two methods close to zero, bias value 3.0263. The error was within the limits of the confidence intervals and these values had a narrow difference. The correlation between the two methods was demonstrated by the linear regression plotted as R2.Conclusion:CellProfiler® image analysis presented similar results to the MTT assay in the identification of viable cells, and image analysis may assist part of biological analysis procedures.
