Objective:The objective of the study was to evaluate the anti-inflammatory and antinociceptive effects of Artemisia afra.Methods: Animals were randomly divided into five groups of six animals each and administered with normal saline (2 ml/kg), indomethacin (10 mg/ kg), and A. afra at doses of 100, 200, and 400 mg/kg, respectively. For the anti-inflammatory activity, carrageenan-induced paw edema was used while the hot plate and acetic acid induced-writhing tests were used to assess the antinociceptive activity.Results: Pretreatment with A. afra at a dose of 100 mg/kg did not show any significant biological effects (p>0.05) for any of the three tests, when compared against saline-treated control group. At a dose of 200 mg/kg, A. afra demonstrated significant effects (p<0.01), during the 5 th h reducing carrageenan-induced paw edema by 12%. The highest dose (400 mg/kg) of A. afra demonstrated more potent effects by decreasing the carrageenaninduced paw swelling (p<0.001-0.05) during the 3 rd , 4 th , and 5 th h, by up to 38% when compared against saline-treated control group. Both the 200 and 400 mg/kg, A. afra doses achieved a significant increase (p<0.05) in reaction time in the hot plate test. In the acetic acid-induced writhing test, pretreatment with A. afra (400 mg/kg) significantly reduced pain by 39% (p<0.01) by comparison with the saline control.Conclusion: Experimental data demonstrated that aqueous extract of A. afra possesses anti-inflammatory and antinociceptive properties in experimental acute inflammation and pain. These findings support the usage of A. afra in managing inflammation and pain in traditional practice.

Anti-inflammatory activity of bartogenic acid containing fraction of fruits of Barringtonia racemosa Roxb. in acute and chronic animal models of inflammation

The fruits of Barringtonia racemosa are traditionally used in Indian medicine for the treatment of pain and inflammatory conditions. In this study, a fraction of ethyl acetate extract of fruits of B. racemosa (BREAF) was investigated for anti-inflammatory activity in experimental models of acute and chronic inflammation. Activity against acute inflammation was evaluated in inflammogens induced rat paw edema models. Whereas, effect in chronic inflammation was evaluated in cotton pellet granuloma and oxazolone induced delayed type hypersensitivity (DTH) model in mice. The BREAF exhibited dose dependent anti-inflammatory activity in both acute and chronic models at oral doses of 5, 10 and 20 mg/kg. BREAF inhibited both phases of carrageenan induced rat paw inflammation. The reduction in paw inflammation by BREAF was also evident in histamine and serotonin induced inflammation in rats. Effect of BREAF on DTH indicates inhibition of immune mediated inflammation. The reduction in cotton pellet granuloma by BREAF treatment shows inhibition of proliferative changes associated with chronic inflammation. Analysis of BREAF after chromatographic separations showed presence of bartogenic acid as a major constituent. Hence, it is proposed that anti-inflammatory effects of BREAF can be partially attributed to its bartogenic acid content. The minute doses at which this fraction shows anti-inflammatory effects emphasizes the need for further investigations on its efficacy in the immuno-inflammatory conditions.

Introduction: Blueberries are rich sources of phenolic compounds and exhibit strong antioxidant activities and a wide variety of biological activities. Methods: Polyphenols were extracted from blueberry by solvent extraction method using ethanol solution. The purification of polyphenols from blueberries (Vaccinium spp.) by macroporous resin XAD-7HP was optimized by adsorption and desorption tests. The cellular antioxidant activities of the purified polyphenols were investigated. Results: The adsorption data indicated that the process was best explained by the Langmuir isotherm model. The best conditions were determined as: blueberry polyphenols concentration, 2.00 mg/g; pH 2.0; ethanol desorption solution concentration, 70 % (v/v); flow rates for feeding and elution, 1 and 2 bed volume (BV)/h, respectively; and a water wash volume of 3 BV. Under these conditions, the purity of the blueberry polyphenols was improved from 46.3 to 86.48 %. The EC50 (concentration for 50 % of maximal effect) and CAA (cellular antioxidant activity) values of the blueberry polyphenols were 151.58 mg/mL and 6.39 µmol QE /100 g sample, respectively.Conclusion: The results demonstrate that purification of polyphenols by macroporous resins XAD-7HP will provide foundation for the development of large-scale separation of blueberry polyphenols in industry.
