Endophytic fungus strain 28 isolated from Houttuynia cordata possesses wide-spectrum antifungal activity

Abstract: The aim of this paper is to identify and investigate an endophytic fungus (strain 28) that was isolated from Houttuynia cordata Thunb, a famous and widely-used Traditional Chinese Medicine. Based on morphological methods and a phylogenetic analysis of ITS sequences, this strain was identified as Chaetomium globosum. An antifungal activity bioassay demonstrated that the crude ethyl acetate (EtOAc) extracts of strain 28 had a wide antifungal spectrum and strong antimicrobial activity, particularly against Exsero… Show more

“…Thus, the current study demonstrated the isolation of endophytic fungus from the rhizome of medicinal plant C. caesia and was identified as O. brasiliensis strain EPE-10 on the basis of nucleotide sequence and morphological characteristics. Previously, several endophytic fungi have been isolated from the medicinally important plants such as Maytenus hookeri (Ni et al 2008), Ginkgo biloba (Qin et al 2009), rhizome of C. wenyujin (Wang et al 2012), Withania somnifera (Kumar et al 2013), Houttuynia cordata (Pan et al 2016). However, to best of our knowledge, there is no report in the literature suggesting the isolation of endophytic fungus O. brasiliensis from the rhizome of C. caesia.…”

“…Santiago et al [ 42 ] also found one endophytic fungal isolate from Cinnamomum mollissimum that possessed efficient killing ability against the pathogenic fungus Aspergillus niger . Pan et al [ 43 ] found that an EtOAc extract of the endophytic fungus Chaetomium globosum from Houttuynia cordata showed a wide antifungal spectrum. The specific secondary metabolites of endophytic fungi, such as helvolic acid, fumitremorgin B, verruculogen, and spirobisnaphthalenes, were also found to exhibit strong antifungal activity against multifarious plant pathogenic fungi [ 44 , 45 , 46 ].…”

“…The total DNA was amplified by PCR using primers ITS1 (5′-TCCGTAGGTGAACCTGCGG-3′) and ITS4 (5′-TCCTCCGCTTATTGATATGC-3′) synthesized by Sangon Biotech Co., Ltd. (Shanghai, China) [ 43 ]. Amplification was conducted in 30 μL of PCR mixture containing 15 μL Premix TaqTM (Takara Biotechnology Co., Ltd., Dalian, China No.…”

“…The present study focuses mainly on targeting significant endophytes from a local variety of rice and maize grown in the Indian region of Indo-Burma biodiversity hotspot with special reference to Manipur having disease control ability, plant growth promoting potential with a broad host range and better option to promote organic farming. Feng pan in his study pointed out the indispensable role of fungal endophyte in biocontrol and renewed attention being paid to the study focussing on the improved host disease resistance mechanism, alongside the secondary metabolites produce by them being a continuous source of new lead compounds and chemical entities in the fields of agriculture and medicine (Pan et al, 2016 ). The opportunity to discover new endophytes with promising properties from this unique ecosystem is appealing.…”
