Abstract: Global DNA methylation was classically considered the relative percentage of 5-methylcysine (5mC) with respect to total cytosine (C). Early approaches were based on the use of high-performance separation technologies and UV detection. However, the recent development of protocols using mass spectrometry for the detection has increased sensibility and permitted the precise identification of peak compounds based on their molecular masses. This allows work to be conducted with much less genomic DNA starting materi… Show more

“…High-performance liquid chromatography and capillary electrophoresis [454,455] methodologies with various mass spectrometry detection and quantification systems [456,457] are the gold standards because they provide absolute measures of GGDm and histone modifications. The strengths of these methodologies reside in the provision of highly accurate data and addition of assay modifications to simultaneously measure other modified bases of epigenetic relevance [458][459][460].…”

“…As a result, in this study, we have tried to evaluate if all those changes can be attributed to the establishment of an epigenetic memory based on specific modifications of cytosine residues by heat. To this aim, high liquid chromatography (HPLC) coupled to mass spectrometry has been employed, which allows a sensible and precise identification of the different nucleotides, including 5mhC, and requires little amount of genomic DNA starting material [ 28 ]. In parallel, the differential expression of putative stress-related genes throughout the SE process has been analysed to assess if the observed epigenetic changes have provoked modifications at the transcriptome level.…”

“…Mass spectrometry is a precise technology for global DNA methylation detection. Usually, nano–ultra-HPLC is coupled to multiple reaction monitoring in a mass spectrometry to achieve the exact quantification of DNA [ 32 ]. Mass spectrometry for global DNA methylation analysis is a highly sensitive and precise method with good reproducibility.…”
